Differentiation of Cord Blood Stem cells into Thymus (T) cells with regulatory phenotype and function. This project will develop technologies for a stem cell therapy platform based on cord blood stem cells, to enable treatment of autoimmune diseases or transplants. Building on the University of Adelaide's frontier demonstration of differentiation of regulatory Thymus (T) cells from cord blood stem cells, the project will develop techniques to expand the numbers of T cells generated. This has the potential to maintain Australia's lead in differentiation of cord blood stem cells and to provide a significant breakthrough in potential treatments of autoimmune diseases (e.g. type 1 diabetes) or transplantation. These diseases affect both a healthy start to life and healthy ageing, and an Australian invention to treat or cure them would have global impact.Read moreRead less

Summary
Adelaide Core Live Organism Imaging Facility. Live organism imaging represents a powerful and essential tool in many aspects of modern biology. This application is for the purchase of two major items of equipment: a Xenogen IVIS Imaging System 200 and a Skyscan 1076_in vivo micro-CT scanner. As there are presently no machines within Adelaide capable of real-time live animal and plant imaging, scientific progress in a number of projects is significantly restricted. The acquisition of a state-of-the-art live organism imaging facility in Adelaide would be a major advance for investigators within the Adelaide bioscience community and would increase their research productivity and international competitivenessRead moreRead less
The role of the neuronal Hu proteins in the regulation of the BMP signalling pathway. We aim to understand the critical decision of a neural progenitor to commit to becoming a neuron. We aim to understand the critical decision of a neural progenitor to commit to becoming a neuron. The BMP signalling pathway is central in this decision. Neural progenitors appear to become insensitive to BMP signals, and this lack of signalling leads to neuronal differentiation. We hypothesise that neuronal identity is regulated by an unusual genetic switch- the translational regulation by the neuronal Hu proteins of two proteins in the BMP pathway. Verification of a post-transcriptional regulatory mechanism for cell fate determination would be a major discovery, and may prompt investigation of how to harness the neuron-inducing function of the Hu proteins to address the therapeutic need for new neurons in neurologic diseases.Read moreRead less
The basis of recognition and disposal of dysfunctional proteins by clusterin. When proteins become damaged they can precipitate. A blood protein called clusterin prevents precipitation of damaged proteins. Clusterin does this by forming complexes with the damaged proteins. Clusterin is the first blood protein known to do this. We will discover which parts of clusterin are responsible for this activity. We will also discover whether cells can take up and dispose of the complexes of clusterin and ....The basis of recognition and disposal of dysfunctional proteins by clusterin. Clusterin is the first blood protein known to do this. We will discover which parts of clusterin are responsible for this activity. We will also discover whether cells can take up and dispose of the complexes of clusterin and damaged proteins. This work is important because some diseases (eg, Alzheimers disease) involve the toxic effects of abnormal protein precipitation. Understanding how clusterin works may help in developing better treatments for these diseases.Read moreRead less
Role of plant host factors in the replication and pathogenesis of Tomato leaf curl virus (TLCV). Knowledge gained from this research will have direct benefit in safeguarding this industry by providing an insight into geminivirus replication and identifying molecular targets for virus control. More generally, the technology developed through this study will contribute to our basic understanding of virus replication in plants with a view to controlling virus diseases by direct molecular intervention with specifically engineered tools. We foresee potential industrial applications. Read moreRead less
Structural studies of the interactions of actinin-4 and intracellular signalling proteins. The actin-based cytoskeleton is one such network of proteins responsible for a number of processes including cell division, migration and adhesion to other cells and tissues. This proposal aims to understand how actinin-4, a component of the actin cytoskeleton in non-muscle tissues, interacts with and is stimulated by proteins of the intracellular signalling cascade.Read moreRead less
Reduced Water Consumption in Commercial Malting Processes. The successful outcome for the project will be the production of barley varieties that can be successfully malted with a single steep, rather than the conventional two steeps currently employed. The objective will be achieved through the reduction of cell wall beta-glucan in barley grain. This will result in water savings of around 40%, or 1,500 megalitres per annum, for the Australian malting industry. This will result in water savings of around 40%, or 1,500 megalitres per annum, for the Australian malting industry. The 1,500 megalitres saving in water use would be sufficient to support the domestic water consumption of 30,000 people, based upon the use of 140 litres per person per day that was recently achieved in Brisbane.Read moreRead less
Development of novel therapies for the treatment of cancer. Both aging and obesity are significant risk factors for cancer and are becoming a burden on the health care budget. The proposed novel cancer therapy will improve current cancer treatments by enhancing their efficacy, thereby reducing the required dose and minimizing side effects. Such an outcome would not only benefit the well being of the individual but would achieve significant health care cost savings.
Determining the regulation of vitamin D metabolism. The proposed project will lead to a better understanding of factors that influence the biological function of vitamin D. This will impact in several areas of human health and will provide new avenues for the development of preventative approaches and treatment of cancer. This project is based on the use of 'Frontier Technologies' that will be applied to elucidate basic biological questions.
