Intravital super-resolution imaging via Stimulated Emission Depletion microscopy (STED)-microendoscopy. We will develop a new technology to enable the imaging of sub-cellular structures within a biological specimen, with super-resolution. This intravital super-resolution imaging technology will build off world leading techniques to image objects with super-resolution and to perform this within a specimen, with minimal invasion. We will develop a new technology to enable the imaging of sub-cellular structures within a biological specimen, with super-resolution. This intravital super-resolution imaging technology will build off world leading techniques to image objects with super-resolution and to perform this within a specimen, with minimal invasion. The broad ramifications of this technology apply to biology, medical science, imaging and sensing. Important applications include the early detection of debilitating diseases and the advancement of understanding of cellular biology. This research will raise Australia's profile as a world leader in science and technology, building on our emerging presence in the biophysical sciences.Read moreRead less
Raman and synchrotron spectroscopy of nano-scale drug interactions and molecular processes in single living cells. The need for potent low-cost drugs is ever increasing, yet effective ways to screen for new drugs remain elusive. A spectroscopic approach to screening drugs in living cells would seem a logical alternative to chemically based and morphological methods that are the status quo. The need for potent low-cost drugs is ever increasing, yet effective ways to screen for new drugs remain elusive. A spectroscopic approach to screening drugs in living cells would seem a logical alternative to chemically based and morphological methods that are the status quo. In this context we are developing methodology to analyse molecular target sites in single living cells for two of the most devastating diseases to afflict human kind, namely malaria and cancer. New ways of rapidly screening drugs in living cells prior to clinical trials will save an enormous amount of time, money and ultimately lives.Read moreRead less
Understanding and exploiting bacterial sulfatases. Bacterial sulfatases participate in environmental nutrient cycling and are implicated in bacterial pathogenesis mechanisms. These enzymes catalyze the hydrolysis of sulfate esters and possess an unusual posttranslational active-site modification where a cysteine residue is oxidized to formylglycine. We will study the mechanism of these enzymes in detail and design inhibitors that exploit the reactivity of this aminoacid. This work has significance because of application to areas that include the treatment of cancer and bacterial infections. Additionally, we will clone novel carbohydrate sulfatases from the heparin-degrading bacterium Flavobacterium heparinum. These sulfatases will have use in biotechnology for characterization of sulfated glycoconjugates.Read moreRead less
