Summary
Cardiovascular diseases kill an Australian every 11 minutes. The goal of this proposal is to characterize the role of the conserved heart regulators in cardiac function and disease. Our combined multi-species and informatics approach has identified a major disease locus for sudden cardiac death in humans, which we propose to characterize. This work can lead to new classes of drugs to improve cardiac health and also aid in early diagnosis of patients susceptible to sudden cardiac death.

Summary
Hormones released from the kidney are important for setting the level of blood pressure. We have discovered that very low levels of the peptide angiotensin in the blood also leads to activation of specific parts of the brain that drive to increase the nervous system activity to blood vessels and therefore increases blood pressure further. This study will explore the chemical signals in the brain mediating this increase in blood pressure.

Cardiac fibrosis is a serious medical condition which can lead to an irregular heart beat and heart failure. We will develop novel diagnostic agents which are more sensitive and able to detect fibrosis of the heart at an earlier time point. This would allow timely initiation of therapy. In addition, we will investigate novel targeted anti-fibrotic agents to treat this devastating disease condition more effectively and with less adverse effects.

Summary
A rise in calcium and free radicals in the heart are associated with the development of heart disease. We have good evidence that a protein in the heart muscle known as the L-type calcium channel mediates changes in calcium in response to free radicals. This proposal will identify how the channel function is altered by free radicals so that a therapeutic target can be designed to prevent altered channel function and development of heart disease during increases in free radicals.

In this project we aim to provide a targeted therapy that inhibits atherosclerosis, in-stent restenosis and thrombosis; pathologies characterized by high shear stress due to a reduction in the vessel lumen. We will apply microfluidic technology to characterize lipid nano-capsules that are tagged with antibodies against activated platelets or VCAM-1, loaded with anti-platelet or immune suppressive drugs and are prone to rupture specifically under high shear stress conditions.

Summary
Acute intracerebral haemorrhage (ICH) is a serious form of stroke. Survivors of ICH are at high risk of repeat events. Blood pressure lowering is a very important to prevent repeat events but data shows blood pressure is poorly controlled in these patients. In this research we investigate whether an approach that uses a 'triple pill' strategy (3 low dose BP drugs in one pill) in ICH patients with mild to moderate hypertension can decrease major cardiovascular events.

Summary
Lifetime risk of coronary artery disease (CAD) is doubled in families with premature CAD. This study will be the first randomized controlled trial of the use of coronary artery calcium scoring (CCS) in relatives of patients with premature CAD, in which treatment will be initiated based on CCS. At three years, the effectiveness of intervention will be assessed on change in plaque volume at CT coronary angiography. The results inform the guidelines regarding evaluation of families with CAD.

Summary
Existing heart failure therapies largely delay heart failure progression rather than reversing the disease. New therapeutic strategies with the ability of improving function of the failing heart are thus greatly needed. The primary goal of this study is to determine whether lipids that are secreted by the heart in a setting of “good” physiological heart growth (as occurs with exercise) can be targeted to restore function of the failing heart.

In a previous study heart ultrasounds in Aboriginal and Torres Strait Islander children frequently showed suspicious but not diagnostic changes. RhFFUS (Rheumatic Fever Follow-Up Study) aims to see if these changes are associated with a higher risk of ARF and progression to RHD. Summary
During cardiovascular disease an inflammatory protein called myeloperoxidase (MPO) becomes abnormally released into the circulating blood and is transported into diseased blood vessels. Our studies show for the first time that increasing circulating levels of MPO promotes both atherosclerosis and aortic aneurysm. This project will study how MPO promotes inflammatory artery disease and test new drugs for their ability to inhibit this damaging protein and protect against cardiovascular disease.
