Summary
Hormones released from the kidney are important for setting the level of blood pressure. We have discovered that very low levels of the peptide angiotensin in the blood also leads to activation of specific parts of the brain that drive to increase the nervous system activity to blood vessels and therefore increases blood pressure further. This study will explore the chemical signals in the brain mediating this increase in blood pressure.

Summary
Acute kidney injury (AKI) is a common cause of ill-health and death. Despite the frequency and seriousness of AKI no new treatments have developed over the past 40 years. While AKI can occur spontaneously it can also develop after treatment with medications, in particular cancer therapies. In this proposal we will explore the effect of new treatments to prevent AKI. We plan to identify new treatments for patients with AKI, with particular relevance to patients receiving cancer treatments.

Older people in hospital are at high risk of adverse outcomes, a vulnerability commonly referred to as frailty. In community-dwelling older people, frailty status can be measured precisely with a Frailty Index score. This research aims to develop a Frailty Index from a standardised comprehensive geriatric assessment instrument used in the routine care of many patients in hospital. This would assist clinicians to identify vulnerable older inpatients, improving their care in the acute setting.
Overcoming The Differentiation Block In Acute Myeloid Leukaemia

Summary
Acute myeloid leukaemia (AML) is an aggressive leukaemia with poor overall survival. About 50% of AML cases have genetic mutations that disable PU.1, which in turn alters the expression of many other genes that cause leukaemia. We have developed new AML models allowing reversible inhibition of PU.1, and have shown that re-engaging PU.1 function causes AML regression. This project aims to understand PU.1 functions in AML and identify rational drug targets for treatment-resistant disease.
AVERT-DOSE (Determining Optimal Early Rehabilitation After StrokE): A Multi-arm Covariate-adjusted, Response-adaptive Randomised Controlled Trial

Summary
AVERT-DOSE is an international, investigator-led clinical trial, developed in Australia but recruiting >2500 patients from 6 countries around the world. In this trial we will establish the optimal model of early mobility rehabilitation for people with acute stroke. The protocols we develop will be suitable for adoption in clinical practice in developed and developing countries and help reduce the unacceptably high personal and community burden of stroke-related disability world wide.
Motivational drive and reward are survival processes that underpin the maintenance of homeostasis. In humans, dysregulation of these brain circuits manifests in disorders such as depression and addictions. Our projects is focused on a key subgroup of neurons in the hypothalamus that modulates reward and motivation. We aim to understand how potent reward experience (e.g. drugs) or stress modifies these circuits and to identify potential entry points for therapeutic interventions.
Very Early Rehabilitation In SpEech: An RCT Of Aphasia Therapy After Stroke

Summary
Aphasia affects one in three stroke survivors. It interferes with talking, understanding speech, reading and writing. Aphasia is devastating. Research suggests that very early aphasia therapy improves recovery of talking and understanding. However, over 70% of people with aphasia do not get very early treatment. This study promotes a “use it” or “lose it” approach to brain recovery and aims to show that very early aphasia therapy is beneficial and cost effective.
Septic shock is a common clinical problem; it is frequently associated with kidney failure that increases mortality. We aim to determine the changes within the kidney that cause it to fail. We will establish whether oxygen levels and blood flow are altered within the kidney, and if blood is shunted through specific blood vessels, reducing flow in critical areas. Importantly, we will determine if clinical treatments used to improve kidney function cause long-term damage by reducing tissue oxygen.

Lmo2 is a transcription factor whose overexpression is a common cause of T-cell leukaemia. This project seeks to identify downstream targets of Lmo2 that cause T-cell leukemia. In addition, the origins and effects of secondary mutations that collaborate with Lmo2 in causing T-cell leukaemia will be determined. This will improve our understanding of how T-cell leukaemia develops and provide new molecular targets for therapy.
