Perinatal exposure to household and environmental toxins may increase asthma and allergic disease risk. Adverse exposures in this critical developmental window may have a marked and prolonged effect on health. A birth cohort of high-allergy risk children will be used to investigate the effect of common chemical exposures on the risk of asthma and allergic disease up to 25 years. Summary
Persistent asthma is a major problem for Australia yet none of the current therapies do more that control the condition. The long-term solution is to prevent asthma from progressing to the persistent form. The major risk factors are: family history, early allergy and recurrent severe lower respiratory infections (sLRI) in the early life. We will conduct a randomized clinical trial to prevent sLRI using a novel bacterial-derived immunostimulant in infants at high risk of developing asthma.

The proposed study will involve the Barwon Infant Study (BIS) cohort of 1074 infants to provide the first systematic investigation of the hypotheses that the epidemic of allergic disease and asthma in many parts of the world relates to the paucity of fermentable fibre in the modern diet, and that the protective effect of fermentable fibre is mediated by changes in the organisms that colonise the gut and the metabolites that they produce.

Food allergy (FA) has risen to epidemic proportions in recent times and Australia has one of the highest rates in the world. FA is most common in children under 5 years, yet adolescents are at the greatest risk of death from food-related anaphylaxis. We don’t know why this is the case. Our research will describe FA prevalence and will assess the factors placing FA adolescents at high risk of severe allergic reactions. Findings will influence policy for management of FA and anaphylaxis.

White blood cells from cancer patients can be modified in the laboratory to react against tumours. These cells can then be given back to the patient, which can sometimes cause cancer regression. However, often the white blood cells lack strength, or they lack the ability to distinguish between tumour and normal tissues of the body. In this project we seek to make stronger and smarter white blood cells that can deliver a lethal hit against tumours without damaging essential organs of the body.

Allergic diseases like asthma, eczema and hay-fever, prevent our children from getting a healthy start to life, and we don’t know how to prevent these conditions. Vitamin D levels may be critical in the development of childhood asthma and allergies, and they can be easily modified! Using a group of 620 children who we have followed for 20 years, we will identify the role of vitamin D levels in the development of allergic conditions, and factors that modify these relationships.
