Summary
The release of the enzyme myeloperoxidase (MPO) within blood vessels is thought to affect their ‘leakiness’ during periods of inflammation, leading to fluid associated swelling (oedema). We propose that MPO produces oxidant chemicals that increase blood vessel leakage. The aim of our work is to study how these chemicals increase vascular leakage by studying the biochemical pathways involved. These studies could lead to new intervention strategies targeting MPO to reduce excessive tissue oedema.

Up until now there has been no simple tests to predict this reliably.The outcome of this project will give us a better understanding of atherosclerosis and provide a simple non-invasive urine test to detect atherosclerosis which would lend clinicians the opportunity to intervene early.

Coronary artery disease (CAD) is a leading cause of death but remains underdiagnosed so better tests are needed. We plan to perform a simple, new blood test in those with and without CAD. Our theory is that blood levels will be significantly increased only in those with CAD. If our theory is confirmed, this test may then be used to better predict those with CAD even before they develop symptoms. It will allow provision of early prevention strategies which will likely reduce complications of CAD.

Coronary stents are wire mesh tubes inserted into cholesterol blockages in heart blood vessels (arteries) thus improving coronary blood flow and alleviating chest pain. Although life saving in acute heart attacks, their value is limited in stable patients and may be associated with severe complications. In partnership with the health department, this project will evaluate how many patients continue to experience chest pain after elective coronary stenting so that can be used more effectively.

The key to effective prevention is understanding of the roles of risk factors in the development of heart disease. These are best defined through the use of large cohort studies, which are those that follow up a group of individuals over time. Statistical analyses are used to develop prediction equations to quantify the effects of multiple risk factors ....Heart disease is the leading cause of death and ill-health in Australia. Accordingly, it also imposes a significant cost burden to the community. The key to effective prevention is understanding of the roles of risk factors in the development of heart disease. These are best defined through the use of large cohort studies, which are those that follow up a group of individuals over time. Statistical analyses are used to develop prediction equations to quantify the effects of multiple risk factors in terms of their contributions to risk of heart disease. The current heart disease prediction equations most commonly used in Australia are based on older overseas studies, such as the Framingham Heart Study. Other than having low relevance to the current Australian population, they incorporate only a limited range of traditional risk factors. A spectrum of new risk factors is emerging. This study aims to develop risk prediction equations for heart disease that are applicable to the current Australian population, using contemporary data from the Melbourne Collaborative Cohort Study. Results from this study will allow the future onset of heart disease to be predicted with accuracy and confidence, which in turn will allow preventive strategies, including expensive drugs, to be utilised in a more effective manner.
