Summary
Blinding eye diseases with genetic contributing factors affecting the retina or light-sensing part of the eye, damage the vision of millions of people worldwide. The vision impairment is irreversible, untreatable, and worsens with age. In this project, we will use technological breakthroughs in genome correction and sequencing, in combination with stem cell and mouse model applications, to develop pathways to treatment of these currently incurable diseases.
Most diseases are much more common in the elderly. The reasons are poorly understood. We have evidence that the most common eye diseases, cataract and presbyopia, are due to the fact that the large molecules that make up the lens do not turnover. In lenses of older people proteins, which make up the bulk of the transparent tissue, become degraded. It turns out that very long lived proteins are quite common in the body. Neural plasticity in older adult human vision. This project aims to expand our understanding of age related changes in brain function, specifically plasticity. The project will increase knowledge of the role of an inhibitory neurotransmitter GABA in visual plasticity. Expected outcomes include new knowledge regarding the regulation of brain function in adulthood, enabling future research and planning for societal benefit to older Australia.
Dynamic ocular imaging: New tools to study neurodegenerative disease. Neurovascular uncoupling occurs when blood supply and energy production is no longer responsive to the metabolic of nervous tissue. Neurovascular uncoupling is thought to be a key mechanism in the development of debilitating neurodegenerative diseases such as Alzheimer’s disease and glaucoma. Understanding lens aging: the molecular basis of presbyopia. Ageing has major consequences in the deterioration of vision, notably, the inability to focus on near objects. The understanding gained through this study of age-related lens changes may allow us to prescribe drugs or diets that alter lens properties and thus delay the need for glasses. This would have great benefits not just in decreased inconvenience of users, but also in cost to the middle-aged and elderly. Refractive errors such as short-sightedness, long-sightedness or presbyopia (age related decline in near vision) are the leading causes of visual impairment in the world. Of these, presbyopia affects almost 100% of the population above 45 years of age. This represents over 40% of all Australians. Glaucoma, the second leading cause of blindness in developed nations, may be slowed if detected early, however recent studies estimate 50% of Australians with glaucoma are undiagnosed. Automated perimetry is used to detect and track such vision loss, but perimetry algorithms perform poorly.
