Dietary selenium supplementation has great potential as a preventative treatment for a range of human health conditions, including cancer, that widely affect the Australian population. However, the adverse effects of such treatments are not fully recognised. This project will increase our knowledge of how selenium compounds are stored and utilised in the body and relate the information to clinical observations regarding dietary intake of selenium and other compounds. The new understanding generated will delineate the conditions for safe intake, so that the beneficial effects associated with selenium supplementation may be harnessed more effectively.Read moreRead less

Porous silicon biosensor for rapid detection of water-borne contaminants. We have recently demonstrated the rapid degradation of porous silicon by certain transition metal complexes known as biomimetic catalysts. The catalysed degradation forms the basis of a new sensor principle where the porous layer serves as matrix, transducer and signal amplification stage. Using this mechanism, we will develop a biosensor for the rapid detection of contaminants (toxins etc.) in water resources. Reservoir water treatment today is reactive rather than preventive because current analysis is slow. Our biosensor can be turned into a field kit to improve water quality management and prevent acts of deliberate sabotage to the water supply.Read moreRead less
Mass spectrometry to reveal the role of lipids in protein misfolding. This project aims to develop an integrated mass spectrometry-based approach to determine structures and dynamics of membrane associated proteins, and discover how lipids cause protein misfolding implicated in neurodegeneration. Lipid membranes may influence protein (mis)folding, aggregation and related toxicity. However, the complex and dynamic nature of the lipid-protein interactions involved means traditional methods cannot ....Mass spectrometry to reveal the role of lipids in protein misfolding. However, the complex and dynamic nature of the lipid-protein interactions involved means traditional methods cannot easily characterise them, and they are poorly understood at a molecular level. The project expects to develop tools to bridge a critical gap in structural biology, while understanding lipid-protein interactions could lead to new therapies. This will ultimately provide benefits such as reduced health costs and better health outcomes.Read moreRead less
Pushing the Boundaries of Multi-modal Biospectroscopic Microscopies. In order to understand the fundamentals of life processes, diseases, and their treatments, it is essential to probe fundamental changes in molecular processes in cells, tissues and whole organisms. Much of our understanding of these processes has involved the introduction of chemical probes for biospectroscopy, but these have inherent problems because the probe can often change the biochemistry that is being probed. This project will push the boundaries of a variety of micro and nano "probe-free" microscopies to provide fundamental insights into these life processes, which could ultimately lead to improvements in the diagnosis, prevention and treatment of diseases.Read moreRead less
The molecular basis of zinc toxicity to Gram-positive bacteria. Gram-positive bacteria are a major cause of infectious diseases in both developed and developing countries. This project will contribute to our understanding of how zinc causes toxicity to these bacteria and facilitate our exploitation of this Achilles heel, by providing new insights into fundamental aspects of microbial physiology.
Novel Biomimetic Nanosprings:Protein-based Elastomer for Engineering Applications. The ability to produce biomimetic elastomeric components with approximately infinite fatigue life offers significant impact on energy consumption and materials usage. In this project, we seek this goal by bio-macromolecular modification and understanding of the unique proteins from a number of different insects that provide the structural basis of novel bioelastomers with outstanding in-vitro fatigue properties. The project will translate the superior in-vivo properties of these proteins to real-world novel bioelastomers for engineering applications. Such functional materials will find potential use in areas such as microelectromechanical devices (MEMS), actuators, artificial muscles, drug delivery vehicles, etc.Read moreRead less
Gas Phase Reactivity of Charged Peptide and DNA Radicals: Fundamentals and Applications. Radicals derived from the "molecules of life", proteins and DNA, play both beneficial (e.g. enzyme catalysis) and deleterious roles (e.g. protein and DNA damage associated with disease). Two electrospray ionisation mass spectrometry approaches have been discovered to generate charged radicals of related models systems (e.g. peptides and nucleobases). The gas phase chemistry of these species is a largely unchartered area! We will examine the fundamental chemistry (unimolecular and bimolecular reactions) of these systems and build upon some exciting preliminary results which suggest potential applications (e.g. as a proteomics tool to sequence and distinguish between leucine and isoleucine residues in peptides).Read moreRead less
