Analysis of trace concentrations of contaminants in food, water and biota is essential for proper environmental and human health protection, and the ability to analyse different isotopes of nutrients will improve our capability to develop techniques to fortify foods with essential micronutrients. The instrumentation will also assist our understanding of the geological processes, climate and environmental change and the formation and location of mineral deposits having economic potential in Australia. By improving forensic identification techniques, the instrumentation will allow identification and tracking of environmental contamination of the food chain and water supplies, and to identify and track criminal and terrorist activity. This project aims to expand the range of applications of nuclear magnetic resonance (NMR) spectroscopy in pharmaceutical research, where NMR spectroscopy is already used routinely for the identification of chemical compounds that bind to protein targets. The techniques developed aim at providing rapid and broadly applicable tools for 3D structure determinations of chemical compounds bound to their protein target, identification of protein-protein interaction sites and characterization of protein motions. The limits of NMR spectroscopy will be pushed to analyse systems of significantly increased molecular weights. The project includes applications to drug targets such as the dengue virus NS2B/NS3 protease.Read moreRead less

New technologies will be developed that are sufficiently rapid and inexpensive to compete with and replace the mutagenesis experiments that biologists usually perform to identify and characterize the functionally important parts of a protein. Nuclear magnetic resonance (NMR) spectroscopy techniques in combination with various selective labelling schemes will be developed with the goal of identification and structural characterization of protein-ligand interactions at increased rates and enhanced accuracy. In addition, the three-dimensional structures of proteins and protein domains of biologically important functions and unknown fold will be determined by NMR. The project aims at techniques of direct impact in pharmaceutical industry.Read moreRead less

One of the methods proposed here is designed to reveal structural information about the binding mode in solution with atomic detail. This knowledge can significantly accelerate drug development. It is otherwise only available from crystal structures that can not always be determined.Read moreRead less

This discovery has opened up new areas for investigation and for the development of new processes and products. Hydrocarbon oils and hydrophobic powders can now be readily dispersed in water without the use of additives. However, the mechanisms involved have not yet been elucidated and it is the aim of this project to understand the process and develop potential commercial applications.Read moreRead less

Enzymes catalyze quite fantastic chemistry under mild physiological conditions. Many special chemical concepts (such as "transition-state stabilization" and "entropy-enthalpy compensation") proposed to explain these powers are unnecessary. Uniquely for a catalyst, these powers are integral to the structure, properties and dynamics of the protein, as constrained and selected by evolution. The question is how do they work? Answering this requires energetic and thermodynamic analysis beyond current experimental techniques, but accessible by computer simulation. We aim to develop a robust toolkit of simulation methods and protocols, blind test them by predicting the mechanism of a new enzyme, with followup experimental validation.

The research will contribute to the development of biomolecular simulation in Australia by demonstrating its potential to complement experiment, and also promote the effective use of APAC (Australian national supercomputer facilities) resources by providing advanced programs and computational protocols for other researchers. It will assist the diffusion of computational biology technology into industrial applications such as rational drug design and protein engineering, as, for example, in our associated Linkage project grant, and provide novel insights into protein engineering and other sorts of design, which transcend concepts currently used in biomimetic chemistry.Read moreRead less
Designer Surfactants for Creation of Emulsion Properties. We propose new approaches to the design of surfactants for stabilising water in oil emulsions. New block copolymer surfactants may be "tailored" for head group structure and a pre-chosen hydrophilic/lipophilic balance at the oil-water interface using the Australian developed RAFT polymer synthesis method and polymeric inorganic species. The proposers have an extensive background in these areas.

X-ray and neutron scattering techniques, developed by us, allow the interfacial structure of the surfactant to be found and the intellectual "loop" closed in the design process. Preliminary screening will be done using the surface balance and bulk emulsion synthesis.Read moreRead less

Our first SPIRT Grant with ORICA Australia Ltd has successfully identified nanostructures in high internal phase emulsions which confer useful stability and robustness on these industrially important systems. The aim now is to ramify those discoveries both scientifically and technologically in the design of emulsion structure using our, now established, techniques. In particular, the objective is to control both nanoscale and mesoscale structure in emulsion formulation, as well as the rheology and stability of emulsion preparations. To do this we plan to use mixed surfactant systems and to study intersurfactant synergic effects on the structure and fluidity of the interfacial material. We believe that the proposed work is unique and that the outcomes will be scientifically novel and also valuable for Australian industry.Read moreRead less
