All-in-vitro engineering and single molecule analysis of protein complexes. The production and engineering of proteins are key methodologies in life sciences. The current project aims to develop new approaches to accelerate the production and analysis of proteins and to apply them to increase our understanding of the basic mechanisms of cell self-maintenance.
Single molecule spectroscopy-guided design of thermostable industrial enzymes. The production and engineering of proteins are key methodologies in life sciences. The current project aims to develop new approaches to accelerate the production and analysis of proteins and to apply them to discover improved proteins for use as feedstock supplements.
High-throughput microfluidic approach to mapping hierarchies of interactions in the gene regulation machinery. The exploration of protein-protein interactions networks is becoming an extremely active area of research in life sciences. The current project will develop new approaches to accelerate the discovery of novel interacting proteins participating in gene regulation, in order to understand how cells differentiate into different tissues and organs.
Proteomics Analysis of Interactions Between Chaperonin 10 and Cell Surface Proteins. Specific interactions at the cell surface may induce downstream biological responses. In the case of chaperonin 10, Cpn10, interaction at the cell surface results in immunomodulation of the inflammatory response. Cell surface proteins that interact with Cpn10 have not been identified. This project will use chemical crosslinking and proteomic techniques to identify cell surface proteins that interact with Cpn10 a ....Proteomics Analysis of Interactions Between Chaperonin 10 and Cell Surface Proteins. Specific interactions at the cell surface may induce downstream biological responses. In the case of chaperonin 10, Cpn10, interaction at the cell surface results in immunomodulation of the inflammatory response. Cell surface proteins that interact with Cpn10 have not been identified. This project will use chemical crosslinking and proteomic techniques to identify cell surface proteins that interact with Cpn10 and structural features of Cpn10 involved in these interactions. These findings will providde leads for the development of immunomodulatory therapeutics based on Cpn10 interactions. Analytical technologies will be developed that are applicable to other interacting protein systems.Read moreRead less
Single-molecule optofluidics: streamlining high-throughput engineering and analysis of proteins and protein assemblies. This project aims at creating novel technologies for high-throughput engineering and analysis of proteins with single-molecule sensitivity. The platform will considerably accelerate the generation of protein-based diagnostics, new vaccines and therapeutics; it will foster collaborations with industry putting Australia at the forefront of protein research.

Facility for multidimensional fractionation of complex biological mixtures. Acquisition of multidimensional fractionation equipment will allow researchers to separate proteins from complex mixtures, and to compare whole protein profiles of multiple samples. This will permit correlation of specific protein changes associated with infection or disease, a major focus of post-genomic programs of research. The equipment will also provide identification of the key differentiating proteins using mini ....Facility for multidimensional fractionation of complex biological mixtures. This will permit correlation of specific protein changes associated with infection or disease, a major focus of post-genomic programs of research. The equipment will also provide identification of the key differentiating proteins using minimal material. Numerous world-class projects and researchers will be able to move more rapidly and reliably from crude cell extracts to identifiable markers, and maintain their competitive positions the recognition of key targets in drug design, disease diagnosis and vaccine development.Read moreRead less

The complete DNA sequence (the genome) is now known for many organisms and advances are being made to identify the complement of messenger RNA (the transcriptome) and the resultant collection of proteins (the proteome). The genome is largely fixed while the transcriptome and proteome differ between cell types in an organism and constantly vary to adapt the cell to changing conditions. The mediators of these variations are proteins, interacting with each other and with signal molecules. The next frontier in molecular biology is to identify and quantify these protein interactions. Our two institutions have a very large cohort of biologists whose research on proteins would be greatly facilitated by the Biacore 3000 and the ISS K2.Read moreRead less
