Summary
Cardiac dysfunction is common in epilepsy, and could be an important contributor to the increased risk of sudden death in people with epilepsy (SUDEP). In this grant we will investigate: when changes in the cardiac function develop in relation to the epilepsy; if people with chronic epilepsy have similar changes; and what effect seizures and epilepsy has on the nerves innervating the heart. The outcomes have the potential to motivate new treatments and prevention for this important problem.

Parkinson’s Disease occurs in about 1% of the population. It is a distressing, progressive disease that places a severe burden on patients, families and carers. 80-90% of Parkinson’s Disease patients have constipation which causes substantial distress to patients, their families and carers. We have a rational path to therapy for constipation of Parkinson’s Disease that will be pursued in this project
Investigating The Mechanisms That Increase Nerve-evoked Vasoconstriction Following Spinal Cord Injury

Summary
People with spinal cord injury not only lose control of their arms and legs but also lose control of their bladder and bowel. They also have poor control of blood pressure and an overfull bladder or bowel can lead to dangerously high blood pressure. In this project, we are investigating how this abnormal high blood pressure is generated. The aim is to develop treatments which target the mechanisms which increase the blood pressure responses elicited by the bladder and bowel.

Summary
A major complication of pelvic surgery is loss of bladder control, mainly due to nerve injury at the time of removing cancerous tissue. This has a big effect on quality of life. Very little research has been conducted on injured bladder nerves. In this project we will investigate what happens to bladder nerves after injury and how we can make them regrow. We will also investigate if undamaged bladder nerves can be made to compensate for the lost function.

Summary
Obstructive sleep apnea (OSA) is a common sleep disorder which significantly impacts daytime functioning leading to excessive sleepiness, and problems with attention and thinking. Currently, the causes for cognitive impairment in OSA (including attentional lapses and performance deficits) are poorly understood. In the awake state, groups of neurons can briefly go “offline” as they do in sleep. These periods of “local sleep” may explain impaired task performance in OSA.

People in pain move differently, yet the physiological basis for altered movement in pain is poorly understood. This lack of understanding has led to treatments for persistent pain that target generic symptoms with limited effect. The current study is the first to examine how different aspects of the nervous system are altered in pain and how this relates to movement. This information will guide the development of new treatment strategies for persistent pain in future.

Schizophrenia is a severe mental illness, with sub-optimal treatment outcomes. New drugs are being designed to provide more effective treatment. To understand the effects of these drugs, we need to know which brain cells have the protein these new drugs target and which cells are affected in schizophrenia. Our project will determine the localisation of this drug target; this knowledge will help the development of better treatments for schizophrenia.

Summary
Our research aims to identify how specific brain cells detect changes in glucose levels and how ageing and diet affect their function. We identified a subset of nerve cells that detect changes in glucose and the “hunger” hormone ghrelin, their ability to do so adapting with age and nutritional status. This project will investigate the potential of these nerve cells as targets for therapeutic and diet- intervention strategies to target obesity, diabetes and promote healthy ageing.

Although the auditory cortex is key to our understanding of several neurological conditions, including language impairments, the functions of many of its areas are still unknown. Using an animal model, we will examine the roles of different auditory areas in separating important sounds from noise. This is a critical role in coordinating our body’s responses to acoustic stimuli. Schizophrenia is a serious and debilitating psychotic illness often characterized by delusions: fixed, false beliefs that preoccupy the patient and affect behaviour, and which are resistant to current drug treatments. This project investigates dysfunctions in belief mechanisms that allow delusions to form and be maintained. This will help clinicians design more effective programs of cognitive behavioural therapy for psychosis by allowing more focussed interventions to reduce delusions.
