The control of chromosome division during female meiosis. Mammalian eggs are stored life-long and finally mature in the hours before ovulation. This project examines how the chromosomes in the egg are separated properly so as to produce a mature egg capable of being fertilized by a sperm. Often in eggs chromosome division is imprecisely executed, and this project will help us understand why this occurs.

This project aims at combining molecular genetics, transcriptome and proteome analysis to identify genes and pathways underlying high light stress tolerance in previously isolated mutants of the chlorophyte Chlamydomonas reinhardtii. Comprehensive profiles of transcriptome-proteome linkage will be constructed without the complications of multicellularity for this unicellular photosynthetic model organism. We will establish a public proteome reference database and provide new microarrays and molecular markers beneficial for research in Chlamydomonas. We expect to advance understanding of high light resistance mechanisms so that it will eventually be applicable to improve productivity in crop plants growing under various environmental stress conditions.Read moreRead less

Lymphatic vessels play crucial roles in tissue fluid homeostasis, immunity, and fatty acid transport. Despite our recent understanding of genetic pathways that modulate lymphatic cell fate specification, how cellular changes mediate morphogenesis of the lymphatic tree remains to be elucidated. This study will combine cell biology and developmental genetics approaches using mouse and zebrafish transgenic lines that label lymphatic endothelial cells to investigate the role of caveolae proteins in the construction of the lymphatic vascular network. This project aims to improve our fundamental understanding of the processes that govern vascular system assembly and will broaden basic knowledge of organ morphogenesis. Read moreRead less

Mouse models for the identification of factors involved in muscle adaptation. The ability of muscle to adapt to meet functional demands is essential for mobility in normal daily life, in ageing well, in individuals with muscle diseases and nerve damage and in athletes. The ability of muscle to change its cellular composition is desirable for the livestock industry. Knowledge of how genes in muscle cells are regulated to adapt to demands has significant implications for public health and economic benefits. We have devised model systems that will allow us to identify the regulators of these genes in order to develop therapies to combat these changes in ageing and damaged muscle, to improve the quality of meat and optimise sport performance.Read moreRead less

Genetic transformation of the circular genome of the plastids provides a containable means for modifying plant growth by manipulating photosynthesis. Although the transformation mechanism is precise, predicting the level of foreign gene expression is difficult because the amounts of messenger RNA and protein produced by foreign genes in plastids varies widely, even when the protein assembles without difficulty. This project will devise strategies for controlling this variability that will facilitate attempts to exploit plastid transformation for transplanting better versions of the photosynthetic CO2-fixing enzyme, Rubisco, into plants to improve their growth efficiency in terms of water, fertiliser and light use.Read moreRead less

Plant genomics has direct benefit to crop improvement, especially as focussed in the applicants' laboratories. Thus, the Australian agri-food sector will benefit substantially from the acceleration in plant functional genomics that will arise from the installation of the robotics equipment described in the current application, by both underpinning more applied research and also being used directly in crop improvement programs such as are based at the Waite Campus. The outputs will include crops with increased tolerance to biotic and abiotic stresses, a reduced dependence on chemical inputs such as fertilisers and improved food quality, with consequent benefits to the environment and human health and nutrition.Read moreRead less
