Summary
Approximately 46% of cancer patients receiving chemotherapy will experience cognitive impairment. The development of this condition may be linked to another common gut side-effect of chemotherapy- mucositis. The treatment of mucositis pain by potent painkillers called opioids may also increase the risk of cognitive change. This project will determine the nervous system changes occurring in mucositis to identify targets for drug intervention to prevent development of cognitive decline.
Optimising Exercise Prescription For Brain Health In Older Adults At Risk Of Dementia

Summary
To reduce dementia burdens in the community, cost effective and targeted early regenerative strategies are critical. Engaging in frequent aerobic exercise is one strategy that can delay the onset and slow the progression of dementia. However, prescription is limited by an incomplete understanding of how exercise positively influences brain health. Here I will investigate the influence of current exercise levels, intensity and exercise environment on brain health in adults at risk of dementia.
IRAP inhibitors are currently being developed as a new class of drugs for treating dementia and other forms of memory deficits. However, there are still gaps in our knowledge about how these drugs act to improve memory. The experiments outlined in this proposal will provide important insights into the drug action in different mouse models of memory deficit.

Although there have been significant clinical advances in the management of injury and the control of acute pain following tauma, many people still develop disabling conditions of chronic pain. Chronic pain and disability occurs even though the acute signs of trauma have subsided and injuries have healed. People with chronic pain conditions not only experience ongoing changes in sensation (ie., most commonly lowered thresholds for pain, touch evoked pain and spontaneous pain), they also endure a ....Although there have been significant clinical advances in the management of injury and the control of acute pain following tauma, many people still develop disabling conditions of chronic pain. Chronic pain and disability occurs even though the acute signs of trauma have subsided and injuries have healed. People with chronic pain conditions not only experience ongoing changes in sensation (ie., most commonly lowered thresholds for pain, touch evoked pain and spontaneous pain), they also endure a number of disabilities for example disrupted family and social relations, disturbed sleep, loss of appetite, weight changes, loss of sex drive, changes in menstrual cycle, the inability to cope with stressors, and often moderate to severe anxiety and depression. The proposed research aims to (i) identify changes in brain circuits which are responsible for producing these patterns of pain and disability following injury and (ii) attempts to selectively reverse some of these disabilities by reversing the brain changes. Persistent exposure to stressful events can produce serious and lasting disturbances in cognitive function. Our research group has recently identified that microglia may play a very significant role in these disturbances. The studies to be undertaken in this proposal will provide fundamental knowledge on how microglia contribute to neuronal plasticity, and how microglia via their effects on neurons regulate complex cognitive behaviour.

Summary
This research program aims to study two disabling symptoms of multiple sclerosis namely upper limb shaking, or tremor, and memory dysfunction. The first project will develop a new treatment for MS arm tremor, Botulinum toxin injections, into routine clinical practice. The second project aims to make available a computerised test of memory that can be done in clinic waiting rooms or at home. This will help neurologists to rapidly pick up changes in memory in a person with MS and improve care.
