Summary
Haemochromatosis is a preventable genetic iron overload disorder. Untreated, it can shorten life due mainly to liver cirrhosis and cancer. It can be prevented by blood donation to maintain normal iron levels. It is unclear, however, whether treatment is necessary when individuals have moderate elevation of iron in the body. This research project will study the effects of treatment in this group by assessing a number of scans, questionnaires and blood tests in treated and untreated individuals.

A successful transition to school sets a child on a path for life. A poor transition can lead to early drop out, poor peer relationships and poor emotional skills. In 2008/09, we found that a brief sleep intervention for children improved key determinants of a successful transition including social-emotional functioning and parent mental health. We now plan to see if the same intervention, delivered by school nurses, can have a similar effect.

Transient ischaemic attacks (TIAs: self-limited brief episodes of stroke symptoms) are high-risk events with an approximately 15% risk of stroke within 90-days. Current practice calls for specialist treatment within 1-7 days. But this recommendation (based on 20 year old overseas evidence) may not apply in contemporary Australia. We will conduct a study of patients with TIA that will document their interactions with Australian health systems and inform contemporary Australian practice.

Learning difficulties are common and can cause school failure and poor self-esteem. They are associated with specific problems with temporarily remembering and using information (‘working memory’). Research suggests that improving working memory might improve academic achievement. We will study this intervention in a large group of primary school children who have poor working memory. If successful, the intervention will provide a way to improve the learning skills of these high-risk children.

In the last decade, there have been reports of powerful antidepressant effects after a single injection of anaesthetic ketamine, with dramatic (though shortlasting) effects within 24 hours. This will be the first controlled study to test whether a course of repeated ketamine treatments, given over 4 weeks, is effective and safe in treating depression.
Antibiotic resistance is a looming public health crisis. New antibiotics with new mechanisms of action are desperately needed. The long-term goal of this research is to develop new drugs that disarm bacteria to overcome the problem of antibiotic resistance.

The ASPREE (ASPirin in Reducing Events in the Elderly) study is a 5 year RCT of aspirin (100mg daily) or placebo in 19,000 healthy people over 70. We aim to augment the existing infrastructure of ASPREE in order to confirm the utility of aspirin for the prevention of depression in the elderly (ASPREE-D). The primary aim of ASPREE-D is to determine if use of low-dose aspirin reduces the incidence of de-novo episodes of depression in healthy individuals over 70 years of age.

There has been an increasing emphasis on intervening early in psychotic disorders. A fundamental principle in early intervention is “to do no harm” and benefits must outweigh the risks of treatment. While antipsychotic medication is very effective and evidence-based form of treatment for positive symptoms in most first episode psychosis (FEP) patients it has risks. This study has the potential to determine whether antipsychotic medication should be the initial treatment option for FEP.
