The normal development of an embryo depends on complex and finely tuned gene regulatory mechanisms. In this Fellowship, I will use sophisticated new technologies to discover which of our 30,000 genes is important for embryonic development, reveal the roles of these genes, and identify the control mechanisms that can go awry to cause birth defects. Our research will suggest new ways to diagnose and deal with these conditions, and will be applicable to stem cell technologies, tissue regeneration, cancer biology, conservation, pest management and livestock breeding, thus delivering significant economic and social benefits to Australia. Australian sugar exports generate almost $2 billion in annual sales, making the sugar industry a critical facet of the Australian economy. In 2006, Australia's primary sugar producing region came under threat when an outbreak of sugarcane smut caused by the fungus Ustilago scitaminea first appeared in Queensland. Management of this potentially devastating disease has focused on breeding programmes aimed at developing resistant sugarcane cultivars, a complex process hampered by a lack of information about the mechanisms of smut resistance. Our research will provide key insight into the mechanisms by which U. scitaminea infects sugarcane, directing future breeding efforts and protecting this valuable industry against further outbreaks.Read moreRead less
Half a Genome is Better than None: The Evolution of Haplodiploidy in Mites. It is our normal experience that males and females are about equally common. We also expect both sexes to receive half their genes from each parent. In many successful animal groups, however, these normal expectations are not met: males are rare and have only half as many chromosomes as females (haplodiploidy) or are non-existent. We will investigate multiple origins of haplodiploid and all-female genetic systems in a ....Half a Genome is Better than None: The Evolution of Haplodiploidy in Mites. In many successful animal groups, however, these normal expectations are not met: males are rare and have only half as many chromosomes as females (haplodiploidy) or are non-existent. We will investigate multiple origins of haplodiploid and all-female genetic systems in a group of mites that includes many useful biocontrol agents and important parasites. The implications of this research have both pure scientific merit and potential practical value in controlling the reproduction of pests.Read moreRead less
Understanding the molecular basis of marine invertebrate larval settlement and metamorphosis using complementary molecular, developmental, chemical and ecological approaches. The colonisation and fouling of submerged structures is a major economic problem for maritime industries. This proposal will detail the molecular mechanisms underlying the first crucial steps of marine invertebrate colonisation - settlement and metamorphosis - and determine how natural anti-foulant chemicals impact on these developmental processes. By using complementary developmental, molecular, chemical and ecological approaches, we will obtain knowledge of key control points in settlement and metamorphosis, and thus identify potential pest management strategies. This proposal comprehensively delineates the role of novel anti-foulants, linking their effect on invertebrate development with their role in shaping marine communities on natural and built surfaces.Read moreRead less
Actin cytoskeleton regulation by E-cadherin and Src. This project examines a fundamental, novel mechanism of how cells work together in tissues. It will provide important new knowledge about how tissues become organized in health, and how organization might be disturbed in disease. It will build Australia's skill base in cutting-edge scientific research, and promote knowledge directed to the research priority area of Promoting and Maintaining Good Health.
How the Y Chromosome makes a male: Molecular genetic analysis of key sex-determining genes. Sex reversal and intersex syndromes are among the most common and highly stigmatized disorders affecting newborn babies. Our research will reveal how the Y chromosome regulates normal male development, identify the steps that go wrong in many male babies, and suggest ways to diagnose and deal with these conditions. It will also pave the way for biotechnological applications in the areas of stem cell techn ....How the Y Chromosome makes a male: Molecular genetic analysis of key sex-determining genes. MOLECULAR GENETICS OF MAMMALIAN SEXUAL DEVELOPMENT: Molecular roles of SRY and SOX9. The development of sexual characteristics is critical to the survival of almost all animal species. This project seeks to clarify how male and female embryos develop differently, focusing on the Y-chromosome maleness gene Sry, and a closely related and equally important gene Sox9. We will study how these genes are switched on in developing gonads and how they interact with other genes to bring about testis forma ....MOLECULAR GENETICS OF MAMMALIAN SEXUAL DEVELOPMENT: Molecular roles of SRY and SOX9. Enhanced genetic analysis facilities at James Cook University. The acquisition of molecular data, DNA sequences or allele frequencies, is the major requirement of almost all laboratory research in the life sciences, and is the rate-limiting step for many research projects locally. The current application underpins novel research initiatives at James Cook University, and is intended to provide centralized state-of-the-art high throughput DNA sequencing and genotyping facility for the north Queens ....Enhanced genetic analysis facilities at James Cook University. Early life-history sex determination in reef fishes. In many fishes, gender is influenced by environmental conditions experienced after hatching. However, the timing, cues, and developmental pathways of sex determination are poorly understood. This research will establish how environmental conditions during the juvenile phase control patterns of sex determination in reef fishes and determine if sex differentiation is a response to local- or broad-scale environmental cues. However, the timing, cues, and developmental pathways of sex determination are poorly understood. This research will establish how environmental conditions during the juvenile phase control patterns of sex determination in reef fishes and determine if sex differentiation is a response to local- or broad-scale environmental cues. An individual's gender has profound and significant consequences for all aspects of its biology and the mechanism of sex determination has important implications for population ecology. This project will test theory in evolutionary ecology and greatly benefit the management of reef fisheries.Read moreRead less
