Summary
Infants with asymmetric brain lesions are at high risk of congenital hemiplegia. This study compares modified CIMT to an equal dose of bimanual training in 150 infants recruited at 3-6 months. Both therapies will be parent-delivered supported by experienced clinicians. Outcomes include use of the impaired hand in bimanual tasks, cognitive and motor development at 12 and 24 months c.a. with measures of neural structure and functional connectivity at 24 months. Early diagnosis of melanoma remains extremely challenging. Currently there are no validated blood-based biomarkers for early diagnosis. Therefore, a reliable screening test is an unmet medical need. Autoantibodies are emerging as promising biomarkers for early cancer detection. In a proof of principle experiment we identified five autoantibodies that provide 95% sensitivity / specificity. Now we will confirm and validate our findings and develop a clinical test for melanoma diagnosis.

Summary
The sequencing of the human genome brings the possibility of more accurate identification of the underlying basis of many diseases. This technology has moved so rapidly, however, that clinical access has been limited. In this application, a national alliance of clinicians, researchers, health economists and policymakers will evaluate the case for clinical genomics across inherited disease and cancer, determine how best to deliver this to the patient and train a capable workforce.
Acute rheumatic fever (ARF) and rheumatic heart disease (RHD) are a major problem for Australian Indigenous people. Accurate diagnosis of ARF allows for early preventive therapy with long acting penicillin, which can prevent RHD from developing or worsening. The aim of this study is to recruit a cohort of children and young people with ARF and apply the latest technologies to their blood samples, with the aim of developing a diagnostic test and open leads for potential new treatments.

Summary
Cerebral Palsy (CP) is common and disability can be progressive so the heathcare burden is immense (0.14% GDP). Our Clinical trials network will improve early detection and develop new interventions to improve physical, cognitive and health outcomes for children with CP and their families. Recruitment from the national CP Register will address clinically important questions and test implementation of effective treatments. New Clinical Practice Guidelines will ensure translation internationally.
