This project aims to characterise key components of nephrons, the glomeruli and tubules, using magnetic resonance imaging without contrast agents, in combination with Deep Learning and super-resolution techniques. Nephrons, the basic functional unit of the kidney, are critical to the maintenance of the body’s homeostasis. Their number and architecture are critical determinants of kidney function. The expected outcomes are innovative semi-automated nephron visualisation and quantitation tools that enable efficient renal phenotyping. Techniques tailored to widely accessible preclinical research scanners are expected to accelerate research into genetic and environmental factors affecting kidney microstructure in embryonic and post-natal life.Read moreRead less

Antisense molecules represent a revolutionary drug discovery platform for life science, but to understand their distributions in cells and tissues is challenging. By integrating nanobiotechnology approaches, this project expects to develop and apply innovative imaging workflow to track antisense molecules in cells and tissues with nanoscale precision. Expected outcomes include new knowledge of the trafficking of these molecules across cells and tissues and refined imaging methods. This project should provide more strategic delivery of antisense molecules to specific cells and tissue, which will have significant downstream economic and social benefits to the Australian community. CRISPR-based pathway activation for bioactive molecule discovery in fungi. Fungi produce an incredible array of unique bioactive molecules, many of which have contributed greatly to humanity (e.g. the antibiotic penicillin, which has saved millions of lives since its discovery). DNA sequencing has revealed many fungi contain the genetic instructions to produce new molecules that have not been seen previously. However, these genes are “switched off" by default and cannot be accessed. This project ....CRISPR-based pathway activation for bioactive molecule discovery in fungi. This project will develop innovative new methods to "hot-wire" these genes, allowing them to turn on and produce a treasure trove of new bioactive molecules. The outcomes of this project will transform our abilities to tap into the hidden potential of fungi to generate new lead molecules for the agricultural and medical industries.Read moreRead less

DNA methylation (mC) is an epigenetic signal essential for the maintenance of correct gene expression patterns. To investigate the causal relationships between mC and transcription during vertebrate embryonic development and evolution, this project will perform high-resolution mC profiling at different stages of teleost, amphibian and mammalian development. Highly conserved and syntenic, methylated sequences will then be used as baits in proteomics screens to identify novel 5mC 'readers'. The generation of genomic profiles of mC 'readers' and their integration with developmental mC maps will reveal transient epigenome dynamics during vertebrate embryogenesis and provide new insights into the conservation of these crucial developmental mechanisms.Read moreRead less
