Funding Amount
$512,256.00
Summary
Osteoarthritis is a painful and debilitating cartilage disease affecting just under 1 in 10 Australians and costs the Australian economy roughly $12 billion per year. This project will develop computational models of cartilage with the ability to incorporate genetic and environmental risk factors into a predictive model of cartilage disease.

One of the common biochemical characteristics of cancer-leukaemia cells is augmented activity levels of enzymes called tyrosine kinases. A major group of tyrosine kinase involved in several cancer-leukaemia types is called the Src family of tyrosine kinases. One member of this family called Lyn has been our focus of study for several years, investigating the signalling pathways that it is involved in. This molecule has also been implicated in several specific leukaemia (Chronic Myeloid Leukaemia and Acute Myeloid Leukaemia) as well as cancer (Prostate, Colon, Breast) in recent years. We have identified a novel mechanism of down-regulation of this enzyme mediated by an adapter molecule called Cbp, which recruits the Lyn inactivating molecules Csk-Ctk as well as SOCS-1; together they inhibit the activity of Lyn and degrade the enzyme. Using our knowledge of the essential interaction elements of Cbp we will design and test various mini-Cbp molecules for their ability to inactivate and degrade Lyn in leukemic and cancer cells. These molecules may allow us to develop novel therapeutics capable of inactivating-degrading specific tyrosine kinases in cancer and leukaemia.Read moreRead less

Environmental risk assessment of acid sulfate soil formation and pollutant generation in Swan Coastal Plain. The urgency and importance of securing water sources for human use in a sustainable manner is dictated by drying climate and rapid population expansion in Australia. Dropping groundwater levels (less recharge, increased abstraction, enhanced drainage) result in formation of acid sulfate soils in oxidised layers that contain acid-bearing minerals. This project will produce an environmental risk assessment framework as a basis for (i) educated decisions regarding land development and soil disturbance vs conservation as well as (ii) identifying areas suitable for water abstraction from groundwater resources. This will ensure sustainable use of precious groundwater resources in this drying continent of ours.Read moreRead less
Optimizing mine floor ripping strategies for diverse regolith types to enhance rehabilitation success. The aim of the project is to identify poorly structured subsoils developed in regolith materials in rehabilitated bauxite mines and develop procedures for minimising the effects of poor soil structure on tree growth. Well structured regolith types will also be identified as sites requiring less ripping. Large-scale field trials will be established to examine the impact of ripping on different regolith types. Methods will be developed to enable prediction of regolith structure based on instumented rippers, drill hole data and remote sensing. Greater success of revegetation practices and a decrease in annual ripping costs ($1.3 m) are anticipated.Read moreRead less
Ecological sustainability and changing land use in the Pilbara. Land use change is frequently associated with large changes in key elements of 'sustainability? ? biodiversity, productivity, and water and nutrient cycles. However, in the Pilbara, fires and flooding rains are so frequent that detecting consequences of land use change is frequently confounded by other ?disturbances?. We aim, through controlled field experiments, to disentangle effects of land use change (e.g. de-stocking of rangelands, altered water availability through mining activity or infrastructure) from those of natural disturbance and variability (both spatially and temporally). This research will improve understanding of key areas in resilience of rangelands and of major processes that are critical for sustainable management.Read moreRead less

Expanding areas of dryland salinity now seem inevitable in southern Australia. There is good evidence that productive saltland pastures can be developed on these areas, and there are opportunities to use these pastures for gaining carbon credits. The present study will account for the effects of saltland pasture establishment and management on carbon sequestration and for greenhouse gas emissions from their saline, waterlogged soils. Budgets of carbon sequestration on saltland pastures will be critical evidence for Article 3.4 negotiations under the Kyoto Agreement. Scaled-up estimates of the carbon sequestered will account for site factors, composition of pastures and grazing.Read moreRead less

This project will devise nucleic acid and serology methods for detection of Olpidium brassicae, vector of lettuce big-vein disease (LBVD), during various phases of lettuce seedling production in nursery environments. It will be the first study to develop sustainable and environmentally responsible nursery best-practice protocols for integrated management of O. brassicae in routine seedling production and for management within the lettuce industry. These protocols will ensure that the spread of LBVD from contaminated lettuce nurseries to farms is prevented. It will also be a unique study of the establishment of a disease in new land with different soils and agricultural practices.Read moreRead less
