Summary
This research will explore the effects of dietary protein on genetic damage to cells lining the large bowel and risk of developing colorectal cancer. We will determine the degree and type of DNA damage resulting from increased protein, the cellular response to this DNA damage, whether it increases risk for developing bowel cancer and whether it can be minimised by other foods in both an animal model and humans.

While some women have a hereditary predisposition to breast cancer, for most women a variety of factors are responsible for their disease. One thing that appears to be important as a cause of breast cancer is our diet. There are many components of the diet that may play a role. One important factor is the amount and type of fat that we consume. Just how dietary fat causes an increase in breast cancer is not known. Obesity is an increasing problem in the community and is associated with an increased incidence of high blood pressure, high cholesterol and triglycerides, diabetes, heart disease and cancer. Although conventional dietary advice for weight loss is a low fat high carbohydrate and high fibre diet there is a very strong interest in the media and the general population in alternative diets that feature reduced carbohydrate with the remainder of the diet either protein or a mixture of protein and fat ....Obesity is an increasing problem in the community and is associated with an increased incidence of high blood pressure, high cholesterol and triglycerides, diabetes, heart disease and cancer. Although conventional dietary advice for weight loss is a low fat high carbohydrate and high fibre diet there is a very strong interest in the media and the general population in alternative diets that feature reduced carbohydrate with the remainder of the diet either protein or a mixture of protein and fat. Unfortunately there is very little published data to guide the decisions of health professionals in this area and there is a strong demand for such information. We plan to perform 2 large weight loss studies which focus on high protein diets with one exchanging protein for carbohydrate and keeping fat constant and the second exchanging protein for fat keeping carbohydrate constant. We will assess whether the high protein diets blunt the decrease in metabolic rate that occurs with weight loss, and whether it spares some of the inevitable loss of muscle mass. We will also measure whether high protein diets improve glucose metabolism in people already at risk of diabetes and heart disease because of their obesity and high insulin levels.Read moreRead less

Summary
Kidney disease is a serious complication of diabetes and may occur as a result of a biochemical process known as advanced glycation. These advanced glycation end products (AGEs) accumulate in the kidney causing disruption of function. Due to modern food processing techniques, the Australian diet has a high AGE content. Over-eating foods which are high in AGEs may worsen diabetic kidney disease. This proposal will test the effects of dietary AGE restriction and overfeeding on kidney function.
This proposal is designed to test the protein leverage hypothesis (PLH) in humans: the idea that the level of food consumption in humans, like other animals, is adjusted to maintain a target protein intake. As the prevalence of overweight and obesity increases, with its attendant health problems, the need to identify which dietary components limit rather than exacerbate energy intake is imperative. According to the PLH, the consumption of a diet low in % protein and high in % fat and carbohydrat ....This proposal is designed to test the protein leverage hypothesis (PLH) in humans: the idea that the level of food consumption in humans, like other animals, is adjusted to maintain a target protein intake. As the prevalence of overweight and obesity increases, with its attendant health problems, the need to identify which dietary components limit rather than exacerbate energy intake is imperative. According to the PLH, the consumption of a diet low in % protein and high in % fat and carbohydrate, typical of many Western countries, inevitably requires the ingestion of additional energy to maintain protein intake constant, thus driving weight gain. Conversely, the consumption of a diet that is relatively high in % protein requires the ingestion of lower levels of energy, creating the potential for weight loss. Preliminary experimental and population-level nutritional survey data support the PLH, as does the finding that protein is more satiating than other macronutrients. If, as predicted, small changes in the proportion of protein in diets described in the current study are found to impact on total energy intake there will be significant implications for weight control strategies. Thus, if the PLH is confirmed, public health dietary recommendations and government policy settings for the food industry will need to change. Obesity is largely due to energy intake exceeding energy expenditure. The CI has reported that dietary protein markedly reduces energy intake but the mechanisms underlying this effect remain unknown. The proposed studies will determine whether gastrointestinal mechanisms are involved in the regulation of appetite and energy intake and how they relate to body weight reduction in obese subjects. The research represents a unique approach to the management of obesity and health.

This increase in coronary risk is caused by the cholesterol that is carried in low density lipoproteins (LDL). However, not all cholesterol is bad. A proportion of the cholesterol in blood is carried high density lipoproteins (HDL), which are powerful protectors against heart disease. People with high blood levels of HDL have a significantly reduced risk of developing heart disease. HDL consist of several different types of particles that contain lipids (or fats) and proteins. Not all HDL protect equally against coronary heart disease. Our ability to determine which HDL are the most cardioprotective is limited because it is difficult to separate the different types of particles from each other. One thing we do know is that the cardioprotective properties and metabolism of HDL are influenced by the proteins they contain. A considerable amount is known about the effects of the two main HDL proteins on the metabolism and cardioprotective properties of HDL. However, HDL contain several other proteins which are also important in this regard. This project is concerned with one of those other proteins called apolipoprotein E. The evidence that apolipoprotein E protects against heart disease is indisputable. Despite this, almost nothing is known about its role in HDL metabolism. This is because it is difficult to isolate large amounts of apolipoprotein E-containing HDL from plasma. In order to overcome these problems I have developed a novel method for preparing HDL which contain apolipoprotein E. These preparations are comparable to the apoE-containing HDL in human plasma.
