Summary
Many bacteria, including some which are virulent pathogens such as anthrax (Bacillus anthracis), are able to enter a dormant state by forming spores (sporulation). These spores are extremely robust and may persist in the environment buried in the soil for example for hundreds of years. The initiation of sporulation occurs in response to changes in the cellular and environmental conditions which threaten the free replicating existence of the bacterium. The initiation of sporulation occurs in response to changes in the cellular and environmental conditions which threaten the free replicating existence of the bacterium. The process of sporulation is controlled at the molecular level by a complex signaling relay. It is of course vital for the existence of the organism that control of sporulation is tightly regulated - preventing the onset of spore-formation in any but the desired circumstances. We aim to determine the three-dimensional structures of the molecules involved in this regulated process and how, by interacting with each other, they can pass on the signal to the bacterium to either start or stop the spore forming process. Summary
The ability to reliably diagnose malaria infections is key to both the management of individual patients as well as public health efforts to control the disease. Current Rapid Diagnostic Tests (RDTs) for malaria have unacceptable sensitivity. The project will determine the low sensitivity of current malaria RDTs available on the market and help produce a malaria RDT with higher sensitivity and stability. This will bring great health benefits to millions of people.

Cancer is the leading cause of death in Australia (28.4% of deaths in 2003). HRG has been implicated in controlling important aspects of inflammatory and cancer disease progression. Namely, HRG appears to regulate the formation and clearance of substances known as immune complexes - the primary cause of tissue damage in this disease. Furthermore, HRG may also control the process of cell invasion which is crucial for the migration of white blood cells of the immune system (leukocytes) to sites of inflammation to combat infections, and is also an important mechanism by which malignant tumour cells escape from primary tumour sites and spread throughout the circulation to other sites in the body. It is this process that makes cancer such a deadly disease. This study aims to define how HRG contributes to these important processes. Summary
Receptors on the cell surface allow cells to respond to their environment. We have recently discovered a new pathway for controlling the amount of receptors displayed on the cell surface, errors within which will lead to defects in development and diseases like cancer. We are studying how this new pathway controls the balance between how much receptors are destroyed after being activated and how much are recycled back for re-use.

Summary
The drug Vemurafenib results in good responses in melanoma patients. However, patients become resistant to treatment. We have identified specific receptors that can cause Vemurafenib resistance, which can be overcome by combination treatment with drugs to these receptors. We will assess melanoma patient samples for expression of these receptors which will be highly beneficial for selecting combination treatments to prevent drug resistance and ensure better prolonged outcomes.
