New models as tools for defining mechanisms of microbe survival in the urogenital tract. Bacteria that infect the human urogenital tract can cause serious disease and these infections represent a large cost to the health-care system world-wide. This study will focus on how bacteria survive in the human urogenital tract and this will impact on strategies aimed at preventing and treating these infections.
Urinary continence and incontinence in men: New insight through new technologies. Healthy ageing in men is largely overlooked. Disorders of continence are surprisingly common with increasing age, and are socially debilitating. To understand changes in continence and address healthy ageing it is first necessary to gain an understanding of continence mechanisms in healthy men. Healthy ageing in men is largely overlooked. Disorders of continence are surprisingly common with increasing age, and are socially debilitating. To understand changes in continence and address healthy ageing it is first necessary to gain an understanding of continence mechanisms in healthy men. Continence in women has received wide attention, but anatomy and function differ, as do the challenges throughout life (childbirth vs. prostate disease). Using novel methods we will test a new hypothesis of male continence with the goal to provide new physiological discoveries, new methods, and baseline data upon which strategies to enhance healthy ageing and reduce the economic burden of disorders of continence can be developed and refined. Sulfate is an essential nutrient for cell growth and survival. The kidneys and liver help regulate sulfate levels in the body, by yet unknown mechanisms. Recently, we cloned a gene, Sat1, expressed in mouse liver and kidneys, which may be responsible for body sulfate maintenance. In this study, we will determine the physiological importance of Sat1 in cell growth/survival and in controlling body sulfate levels. We will generate and characterise a Sat1 lacking mouse, study its expression during development and its effects on other genes. We will elucidate how body sulfate levels are maintained and its importance in cell growth/development.Read moreRead less

This project seeks to better understand regulatory mechanisms of embryonic development. A key challenge in biology is to understand how genes act in regulatory networks to coordinate the formation of complex organs during embryo development. Although several genes influence embryo sex determination and gonad development, Sox9 has emerged as a master regulator in this system. This project plans to use new systems biology technologies and transgenic mice to identify the suite of Sox9 target genes and partner proteins. Anticipated outcomes include insights into sex determination, stem cell differentiation, and development of many other organ systems that depend on Sox9 function, providing proof-of-concept for high-throughput analysis of molecular control networks in the embryo.Read moreRead less
Morphological development of the kidney – a paradigm for organogenesis. This project aims to shed new light on how the kidney develops, how normal adult function is established and how congenital kidney malformations occur. It aims to investigate a mathematical pattern that has been discovered in the program of branching morphogenesis which dictates kidney development. By combining cutting-edge genetic approaches, morphological analysis and mathematical modelling, this project plans to explore h ....Morphological development of the kidney – a paradigm for organogenesis. This project aims to shed new light on how the kidney develops, how normal adult function is established and how congenital kidney malformations occur. It aims to investigate a mathematical pattern that has been discovered in the program of branching morphogenesis which dictates kidney development. By combining cutting-edge genetic approaches, morphological analysis and mathematical modelling, this project plans to explore how this pattern is established, how it dictates kidney development and whether it constitutes an ancient mechanism central to organ formation. Project outcomes may improve understanding of fundamental developmental processes and developmental disease and variation. Project findings may also be important for understanding the development of other organ systems.Read moreRead less
The structure and patterning of branching morphogenesis in the developing kidney. This project aims to understand a fundamental developmental process known as branching morphogenesis, which drives the formation of many organs including the kidney, lungs and glands. Understanding this process will be of key importance in understand how our organs form.
Switching on sex: How key mammalian sex-determining genes are activated. Development as a male or female is one of the most important biological decisions made during embryonic life. Although several of the key sex-determining genes are now known, it is unclear how they come to be active at the right time and in the right cells of the gonad to direct sex determination. This project aims to use bioinformatics, biochemistry, cellular assays and powerful genome editing technologies in mice to pinpo ....Switching on sex: How key mammalian sex-determining genes are activated. Development as a male or female is one of the most important biological decisions made during embryonic life. Although several of the key sex-determining genes are now known, it is unclear how they come to be active at the right time and in the right cells of the gonad to direct sex determination. This project aims to use bioinformatics, biochemistry, cellular assays and powerful genome editing technologies in mice to pinpoint essential regulatory elements and understand how they function. In addition to addressing key issues in mammalian molecular genetics and developmental biology, data generated in this project may provide a basis for innovative biotechnologies in pest management, wildlife conservation and livestock breeding.Read moreRead less
The makings of maleness: molecular and cellular functions of the Y-chromosomal sex-determining gene Sry. This project will reveal how the Y chromosome directs male development, identify steps that can go wrong, and suggest ways to diagnose and manage intersex conditions. It will also inform stem cell, pest management, wildlife conservation and livestock breeding technologies.
Understanding self-organising tissues. This project will discover how an organ can form from a mixture of component cells by 'self-organisation'. Understanding of how this can occur, could potentially be applied to the bioengineering of organs from component cells.
