This project aims to generate fundamental knowledge on the origin of diversity in mammalian brain circuits by studying development of marsupials and rodents. The expected outcome is to elucidate how differences in the timing, rate and sequence of development of gene expression, cell differentiation and circuit formation can relate to the origin of key evolutionary innovations in the mammalian brain. The significance of understanding the dynamics of developmental systems that shape complex brain traits includes establishing new developmental paradigms in evolutionary theory, generating new tools to investigate and manipulate brain gene expression in vivo, and the potential discovery of the causes of neurodevelopmental dysfunction.Read moreRead less

The species to be considered are those of significant industrial (e.g. C2F4, C4F8 and CF2) and environmental (e.g. CO2 and H2O) interest, with the cross sections we will measure being crucial to our modelling both the processes involved in plasma enhanced chemical vapour desposition of semiconductor devices and the spectral emission characteristics of planetary atmospheres. The modelling will be conducted self-consistently with our enhanced statistical equilibrium simulation code, which is unique to Australia.Read moreRead less
Safer gene editing tools for Australian livestock and biotech industries. Editing the genome of an organism in an efficient and safe fashion is critical for the livestock and biotechnology industries. While CRISPR-Cas9 has become the method of choice for genome editing, it is known to introduce unwanted "on-target" and "off-target" mutations, limiting its utility. To address this the CI team created a novel genome editing platform technology termed Crackling-CAST that is almost 100% accurate, while retaining the efficiency of the classical Cas9 system. This project will exemplify the capabilities of the novel gene targeting platform in cell types used by the biotechnology and livestock sectors, ensuring its global uptake by these industries and delivering significant economic benefits for Australia. Inter- and Intra-molecular interactions probed by electron momentum spectroscopy. High-resolution electron momentum spectroscopy (EMS) will be used to study bonding (intra-molecular interactions) in complex oriented organic species. This will allow us, in conjunction with density functional theory calculations, to evaluate the basis sets and exchange-correlation (XC) functionals employed in those calculations, thereby potentially enabling quantum chemists to build more physically accurate XC-functionals and basis sets. As chemistry largely occurs in solution, the future will increasingly focus on solvated species. Consequently, we are also initiating an EMS study of the electronic wavefunctions of solvated species with our recently developed "clusters" EMS spectrometer. This will provide the first data on the electronic wavefunctions involved in inter-molecular interactions.Read moreRead less

The project aims to address the emerging technologies associated with the interaction of plasmas with liquids and biological matter, including plasma medicine. The project expects to generate new knowledge on the role of electron-induced processes through the development of complete and accurate sets of microscopic cross-sections for electrons with biomolecules within tissue. This microscopic data will inform new microscopic models for non-equilibrium electron transport in liquids and biological matter, and its coupling to plasmas. The expected outcomes of this project include progress towards the optimisation of safety/efficacy of future generation plasma medicine devices through detailed understanding of plasma-biological tissue interactions.Read moreRead less

The interaction of low-temperature plasma electrons with liquids has served as a reducing agent in various technological applications in water treatment, agriculture, biofuels and medicine. Predictive control of the plasma-liquid interface is essential to unlocking the potential of these applications, and this has been limited by the absence of the relevant non-equilibrium transport theory describing electrons at the plasma-liquid interface together with fundamental data describing electron interactions with liquids. The project will develop a state of the art presolvation electron transport model informed by world first measurements of electron cross-sections for radicals and liquids and apply it to model plasma electrochemistry processes.Read moreRead less
The developmental genetics of major evolutionary transitions: a multidisciplinary investigation of limb reduction and loss in lizards. The five-toed limb is an iconic evolutionary innovation of land vertebrates, yet has been lost repeatedly. This project will use anatomical, developmental and genetic approaches to understand how vertebrates lose their legs, whether limbs can be reacquired, and the degeneration of limb genes after they lose their function (analogous to 'vestigial organs').

Mammals lack microchromosomes, but contain gene-rich regions with similar attributes. We suggest that microchromosomes originated by genome duplication, and evolved into the gene-rich regions of mammalian chromosomes. We will test this hypothesis by comparing sequences and genes in microchromosomes of birds, reptiles and monotremes. This will clarify the origin and evolution of the ?microgenome?, establish its suitability as a model for vertebrate genome organisation, and demonstrate whether microchromosomes are the ancestors of the gene-rich regions of mammalian chromosomes.Read moreRead less
