Chip liquid chromatography-inductively coupled plasma-mass spectrometry: a new hyphenated microfluidic instrument for metallomics. An alliance of two Australian universities and a world-leading scientific instrument company will develop innovative technology that will enable new reliable, sensitive and patient-specific medical tests, as well as being used to probe causes of significant diseases that appear to involve trace levels of metals in the body.

New methods are needed to understand complex cellular processes such as metabolism. This proposal will support the development of methods in metabolite profiling and flux analysis that provide a global view of metabolic networks in cells and complement other profiling approaches, such as proteomics and transcriptomics. The development of these approaches (collectively termed Systems Biology) is essential for maintaining Australia science at the forefront of international efforts (National Research Priority 3; Breakthrough science). This project will also directly contribute to our understanding of metabolism of an important human pathogen and provide training to young Australian scientists.Read moreRead less
Phytochemistry Of Thrips Resistance In Potato And Its Influence On Acquisition And Transmission Of Tomato Spotted Wilt Virus. TSWV causes multi-million dollar losses to the Australian potato industry, which is the countries largest and most valuable vegetable commodity and an important source of income and employment for rural communities. Development of TSWV epidemics relies on the activity of vector thrips acquiring virus from infected plants and transmitting it to healthy potatoes. Deployment ....Phytochemistry Of Thrips Resistance In Potato And Its Influence On Acquisition And Transmission Of Tomato Spotted Wilt Virus. Deployment of thrips resistant potato would greatly assist disease control. This project will determine the efficacy of thrips resistance in reducing TSWV epidemics and identify the chemical basis of resistance, which could be used as a screening tool for potato breeding, and could form the basis of a synthetic chemical tool for thrips prevention in potato and other crops.Read moreRead less
The Impact of Changing Climatic Conditions inferred from the Isotope Abundances of Trace Metals in Global Ice Sheets and Glaciers. In this project Greenland and Antarctic ice-cores more than 3 km long will be used to investigate climatic variation extending back more that 4 complete glacial cycles. Some of these ice-cores include sections of refrozen water formed from sub-glacial Antarctic lake water. This project will use naturally occurring lead and strontium isotopic tracers to fingerprint ....The Impact of Changing Climatic Conditions inferred from the Isotope Abundances of Trace Metals in Global Ice Sheets and Glaciers. In this project Greenland and Antarctic ice-cores more than 3 km long will be used to investigate climatic variation extending back more that 4 complete glacial cycles. Some of these ice-cores include sections of refrozen water formed from sub-glacial Antarctic lake water. This project will use naturally occurring lead and strontium isotopic tracers to fingerprint impurities in the ice, because they have the potential to simultaneously signal the timing and location of past episodes of climate change. This will lead to an improvement in our understanding of the processes that cause these changes.Read moreRead less

Medicinal chemistry is constantly being challenged to develop efficient methodologies for the synthesis of compounds for drug discovery. Following completion of the Human Genome project, the cloning and expression of new proteins will proceed at an accelerated rate. In the absence of biochemical knowledge of the target protein there is a growing interest in techniques that expand the structural and biological diversity of compound libraries. Dynamic combinatorial chemistry is an innovative technology with the capacity for supporting the shift from focussed to diverse compound libraries. This application seeks funding to refine dynamic combinatorial chemistry into an effective drug discovery tool.Read moreRead less
New Methods for Structural Biology in Solution. This project aims to expand the range of applications of nuclear magnetic resonance (NMR) spectroscopy in pharmaceutical research, where NMR spectroscopy is already used routinely for the identification of chemical compounds that bind to protein targets. The techniques developed aim at providing rapid and broadly applicable tools for 3D structure determinations of chemical compounds bound to their protein target, identification of protein-protein interaction sites and characterization of protein motions. The limits of NMR spectroscopy will be pushed to analyse systems of significantly increased molecular weights. The project includes applications to drug targets such as the dengue virus NS2B/NS3 protease.Read moreRead less

New technologies will be developed that are sufficiently rapid and inexpensive to compete with and replace the mutagenesis experiments that biologists usually perform to identify and characterize the functionally important parts of a protein. Nuclear magnetic resonance (NMR) spectroscopy techniques in combination with various selective labelling schemes will be developed with the goal of identification and structural characterization of protein-ligand interactions at increased rates and enhanced accuracy. In addition, the three-dimensional structures of proteins and protein domains of biologically important functions and unknown fold will be determined by NMR. The project aims at techniques of direct impact in pharmaceutical industry.Read moreRead less
Storage of Volatiles in the Mantle Lithosphere: Time-scales, sources and processes. Fragments of the Earth's mantle (xenoliths), transported to surface by deep-seated volcanics, record the injection of fluids through formation of volatile-bearing minerals (mantle metasomatism). Fragments of the Earth's mantle (xenoliths), transported to surface by deep-seated volcanics, record the injection of fluids through formation of volatile-bearing minerals (mantle metasomatism). This project will 1) test the hypothesis that timescales of mantle volatile storage can be determined using noble gas (laser probe) dating techniques, and 2) determine the sources and processes involved in volatile storage, using halogen and noble gas geochemistry. A successful outcome will establish unique methods for dating mantle metasomatic events and determine the relationship between mantle and crust melting episodes, thus providing critical constraints on mantle evolution and Earth degassing models.
