A biological model to understand caveolin-1 and lipid raft function in health and disease. This project will generate a biological model for pathological caveolin-1 action on cell membrane domains called lipid rafts to determine how they trigger chronic diseases such as cancer and diabetes. The tools developed in this project will help Australia find new drug targets for the treatment and prevention of these prevalent diseases.

Mitochondria produce the energy for our bodies. Defects in this process cause mitochondrial disease, which affects at least 1/4300 people. Diagnosis is often inconclusive and few if any effective treatments exist. State of the art CRISPR gene-editing tools will be used to make disease models mimicking the different types of mitochondrial disease. These will be used to understand how mitochondria function, identify new disease genes and develop new drugs.

Summary
Drug combinations in chemotherapy hold promise for more effective treatments and for overcoming drug resistance, but the search for effective combinations is challenging. The combination therapy R-CHOP is often curative for Diffuse Large B-cell Lymphoma (DLBCL). Both cellular drug interactions and evolutionary drug interactions will be quantified in DLBCL cells, to understand the defining features of effective combinations and guide the future rational design of combinations.
