In this project new DNA-based technology will be developed to determine the numbers of specific bacteria in the dental plaque of patients. The project will also use new mass spectrometric analysis techniques for the determination of compounds in gingival crevicular fluid, an exudate from the gums. Strabismus is the pathological misalignment of the eyes associated with loss of binocular vision and is one of the most common human ophthalmological disorders. Patients with comitant strabismus have full eye movements, whereas patients with incomitant strabismus have limited eye movements, which causes the angle of strabismus to vary with gaze direction. This project aims to define genetic contributors to comitant congenital strabismus.

Summary
Bowel cancer screening will be most effective in disease prevention if it is applied proportionately to individual person's risk. Risk-based screening requires a risk calculator to assess personal risk. By utilising existing large, international datasets, I will identify the risk factors specific for different bowel cancer types and incorporate them to upgrade the prediction model that I have developed. This will achieve more accurate risk prediction to enable personalised risk-based screening.
Cultivating numerically significant soil bacteria. The vast majority of soil bacteria have not been able to be studied in the laboratory because they cannot be grown outside the soil. They are therefore termed unculturable. Most of these belong to groups that are completely unstudied. Advances made in the Janssen lab have overcome this impediment to laboratory cultivation of numerically abundant and globally distributed soil bacteria. Summary
Losing function in your arms and legs after an injury (quadriplegia) is a catastrophic event. Quadriplegia also results in obstructive sleep apnoea; a condition where the throat closes repeatedly while asleep causing sleepiness, poor concentration and cardiovascular diseases like stroke. Despite most people with quadriplegia having this disease, the cause is unknown. This project will thoroughly investigate obstructive sleep apnoea causes in people with quadriplegia and test a possible treatment
A novel process for removing phosphorus microbiologically from wastewater. Australia is a water limited continent, and its rivers and lakes are priceless assets, but because of climatic conditions, these are especially sensitive to blooms of 'blue green algae'. Some are highly toxic, and water containing them is unsuitable for most purposes. Current technology for P removal requires constructing complex plant configurations, and most of these operate unpredictably and unreliably. It is traditionally taught that the cause of acute rheumatic fever (ARF) is always infection of the throat with the bacterium group A streptococcus (GAS). However, in Aboriginal communities of the Top End of the Northern Territory the incidence of ARF is the highest reported in the world, yet GAS is uncommonly isolated from the throat. There is further information to suggest that GAS skin sores may underlie many cases of ARF. If this were proven, it would completely alter the traditional view of the cause of ARF, and have important implications for prevention of ARF around the world. Presently, these approaches focus on diagnosing and treating sore throat, but no country has proven that such a program can be successful in substantially reducing new cases of ARF. If it was known that skin infection could lead to ARF, then countries (including Australia) could emphasise the importance of skin health programs. A further benefit of this knowledge would be to influence GAS vaccine development, which presently is largely focused on the prevention of sore throat. A different possibility has recently been raised - that the cause of ARF may not always be GAS, but instead that the related bacteria GCS and GGS may have the potential to cause this disease. Proof of this hypothesis would even more dramatically alter our understanding of disease causation, prevention, and vaccine development. We propose to determine the cause of ARF in Aboriginal communities by regularly swabbing families of people with a history of ARF, and using genetic fingerprinting of the bacteria from the skin and throat swabs. When cases of ARF occur, we will be able to determine the site and type of infection that precipitated the attack. We will conduct a related study in more communities, in which we will swab family members of people with ARF and of control families (without ARF) to determine the bacteria most commonly isolated from ARF families.Read moreRead less
