Roles for quorum sensing and biofilm formation by Vibrio cholerae in resistance to protozoan grazing. This research will benefit Australia through an increased fundamental understanding of how our model bacterium, Vibrio cholerae, survives in the environment. This could lead to the development of strategies that control bacterial biofilms, a significant medical and industrial concern. This project will also be of benefit through the training of postgraduate students in environmental microbiology ....Roles for quorum sensing and biofilm formation by Vibrio cholerae in resistance to protozoan grazing. Environmental influence on evolutionary processes in bacterial populations. Bacterial populations exhibit remarkable adaptive capabilities in many environmental and medical settings. They respond to environmental stress in terms of altered gene expression, but what are the effects on mutation rates and fitness when cells grow at suboptimal growth rates? We plan to test whether the physiological state of an ancestral population is a determinant of a population's subsequent response to selection. ....Environmental influence on evolutionary processes in bacterial populations. They respond to environmental stress in terms of altered gene expression, but what are the effects on mutation rates and fitness when cells grow at suboptimal growth rates? We plan to test whether the physiological state of an ancestral population is a determinant of a population's subsequent response to selection. A simple model system using controlled culture conditions will be used to investigate the linkage between environment, mutation rate and fitness under selection. Demonstration of these linkages will have significant impacts on microbiology and understanding evolution.Read moreRead less

Infections associated with biomaterials incur a high cost in terms of human health and well being, but such infections also increase the burden on the health care system by extending hospital stays and significantly elevating costs. The cost of a catheter-related blood stream infection is around $50,000 for patients in intensive care units. If all medical devices are considered, the cost of related infections is then approximately $20 billion. The technology proposed here has the potential to reduce biomaterial related infection rates, improve health care and reduce health care costs. Read moreRead less
Genetics and evolution of Shigella O antigens. We use genome scale sequencing techniques to sequence 26 O-antigen gene clusters from Shigella. O-antigens are important for their life and pathogenicity. This project will greatly extend our knowledge of the genetic basis and evolution of this important polymorphism. O-antigens are used for typing Shigella and also elicit strong immunity. The molecular data will help establish DNA based typing and vaccine development.Read moreRead less

This project aims to determine the basis for plasmid and membrane vesicle generation and DNA transfer at the cellular and molecular level. Recent discovery of plasmid vesicles, which transfer plasmid DNA between host cells using viral capsid-like membrane vesicles, suggests they may be an evolutionary precursor for virus particles. The expected project outcomes include the first substantive characterisation of membrane vesicles in the phylum Euryarchaeota, how plasmid vesicles are generated and transmitted, and new insights into how viruses may evolve. This may lead to new avenues for preventing viral transmission and supporting development of new and improved applications biotechnology and the safe delivery of vaccines or genes in animals and humans.Read moreRead less

We propose to adapt these structures for use in microbial ecology because this field is one of the least understood areas in biology. The technology we will develop will have far broader uses, and will create new diagnostic tools for monitoring and understanding microbial ecosystems would be invaluable in a number of fields. Examples are medical diagnostics, waste-water treatment, bioremediation, food and agriculture, bioprotection and biodiscovery.Read moreRead less

We seek to establish an Australian first confocal laser scanning microscope with time resolved imaging and fluorescence correlation spectroscopy capabilities. Its advantages include ultrasensitive detection of weak fluorescent emissions against high autofluorescent background by using fluorescence lifetime signatures, with over three orders of magnitude discrimination improvement. The system will also be able to monitor binding of single molecules. These techniques will open new and exciting avenues for interdisciplinary research at the frontier between biological and physical sciences. The microscope will operate within an existing multi-user Optical Characterisation Facility supporting research of an established network of scientists in the Sydney area.Read moreRead less
Defining how bacteriophage shape the biofilm lifecycle of bacteria. Bacteriophages are viruses that infect bacteria and they represent a significant selective pressure that drives the evolution of bacteria. We will study the genetic mechanisms by which genes encoded by a bacteriophage can contribute to increased survival of bacteria in the environment.

Infection with Cyclospora cayetanensis is an emerging disease that causes significant morbidity in human populations. Although prevalent in developing countries, recent outbreaks in the USA confirm it is now emerging into the developed world. The research described here will analyse and characterise the antigens on the surface of the oocyst of C. cayetanensis which will add significantly to our sparse knowledge on this important human parasite. We will also develop the first commercial antibodies and technologies which will rapidly and accurately detect the parasite in human patients and our drinking water supplies, thus securing a global market for Australian technology.Read moreRead less
