Summary
In this project we want to see if we can prevent depression by improving insomnia. We will invite people with elevated depression symptoms to undertake a novel self help program to reduce insomnia, and then see if their risk for developing clinical depression is reduced 9 and 18 months later. As far as we know, this approach has not previously been tried. The merit of the project is that we have tangible symptoms (sleep problems) with which to engage individuals in a prevention strategy.

Self-management of symptoms is an important part of treatment for people with knee osteoarthritis. Footwear influences forces across the knee joint and it is unclear which types of shoes are best to minimise arthritis symptoms. This study will compare the effects of flat flexible shoes to stable supportive shoe styles on pain and physical function over 6 months in people with painful knee osteoarthritis.

Smallpox is one the most notorious diseases in human history. Despite its eradication in the 1970s, human cases of animal poxviruses such as monkeypox virus and the potential use of smallpox as a bioterrorism weapon have called for an improved preparedness of Australia against (re)-emerging poxviruses. This project combines structural biology approaches to understand the complex assembly of poxviruses and provide the basis for the development of broad-spectrum antiviral drugs.

Summary
Following inhalation, respiratory viruses can infect and grow in airway epithelial cells. Although immune cells such as macrophages are also susceptible to infection, this is generally abortive and new viruses are not released. This project will identify proteins induced in macrophages that block respiratory viruses and prevent their spread in the airways. We will also define mechanisms by which some virulent strains overcome this block to grow in macrophages.

People with severe mental illness die 13-30 years earlier than other people, mostly from physical illnesses especially cardiovascular disease. Physical health care services are poor for these patients and need urgent improvement. Our team has developed a process to deliver good physical health care using the Flinders Program and recovery framework. This project evaluates the effectiveness and cost-effectiveness of the process, and shows how health services can deliver physical health care.

The Hippo pathway is a key regulator of tissue growth. It was first discovered in vinegar flies and plays a similar role in mammals. We aim to define the mechanism by which the Tao kinase controls tissue growth by regulating the Hippo pathway. These studies will be performed in flies and mammalian cell culture. Our studies will shed light on how tissue growth is controlled, and have the potential to inform the way that we treat human cancers and tissue growth disorders.

For the brain to function correctly its neurons must be connected correctly. This project will use a novel mathematical approach to understand how growing nerve fibres find where to go in the developing brain. In particular we will use both experiments and computational analysis to understand how the shape of the tip of a growing nerve fibre helps the fibre navigate. This may help us understand the biological cause of many different types of mental disorders.

We discovered and manufactured a growth factor produced uniquely by the egg. We named this growth factor cumulin. It is a powerful regulator of ovarian function and egg quality. This project will study the basic mechanisms of how cumulin works in the ovary. We will then develop an assay to measure it as a biomarker of human egg quality and quantity. New approaches in fertility preservation for cancer survivors will be developed using cumulin.

Diseases, infections and pathologies are common clinical problems of the intestinal lining in both infants and adults. Individuals with these conditions can experience nutritional problems and severe cases result in death. The intestinal lining is generated from a small population of stem cells. In this study we use of a mouse model where the stem cells are marked and will examine what factors regulate stem cells in the intestine with the aim of facilitating intestinal tissue regeneration.
