This proposal aims to reveal how the interaction of nuclear RNA and protein molecules control gene regulation in the face of cell stress. To understand how genetic variation leads to changes in the expression of genes, we need new insights into the fundamental principles underpinning complex gene regulatory systems. Building on the discovery of paraspeckles, novel gene regulatory structures, this project will yield insights into gene regulation that will help fill these knowledge gaps. This will provide a more comprehensive understanding of RNA-mediated gene regulation, and will open up new research opportunities to target RNA based gene regulatory complexes.Read moreRead less
The roles and regulators of new plant cells linked to root transport. Plant genomics has moved to the single cell resolution, allowing precise investigations of previously hidden cell types and cell states that respond to environmental stress and that vary among differentially adapted plant populations. Plant genomics has moved to the single cell resolution, allowing precise investigations of previously hidden cell types and cell states that respond to environmental stress and that vary among differentially adapted plant populations. Here, we will extend our pioneering efforts that have mapped and discovered novel root cell types, to determine their salt and nutrient stress responses, and to elegantly dissect the underling causal genetic variation. The unique cell markers and regulatory networks will be validated with tissue specific and transgenic tools that can work across a host of plant species to reveal adaptive cellular responses to harsh environmental conditions.Read moreRead less

Control points in nitrogen uptake: enhancing the response of cereals to nitrogen supply and demand. Vast amounts of nitrogen fertiliser are applied to cereal crops to maintain yields. By uncovering what limits nitrogen uptake in cereals, this project will provide the scientific basis for improving nitrogen use efficiency and decreasing fertiliser use, with significant economic and environmental benefits.
eGenomics - Next generation biomonitoring of threatened species. DNA is the molecule of life and exists everywhere in the environment as a largely untapped source of information on evolution, biodiversity, and ecosystem health. Our overriding aim is to start mining that information to benefit threatened species. Based on optimized ancient DNA methods, powerful sequencing technology, whole genome analyses, and RNA profiling, we present a novel and holistic framework for genetic biomonitoring. In two parallel model systems we will study corals and reptiles to improve environmental detection while simultaneously obtaining information on their population health. This will foster more efficient conservation of endangered species that are of tremendous importance to our marine and terrestrial ecosystems.Read moreRead less
Deciphering the regulation and function of the epigenome in eukaryotic development and stress response. The epigenome is an additional regulatory code superimposed upon plant and animal genomes that controls how they operate. This project will aim to understand the information encoded in the epigenome and how it changes in development and environmental stress, enabling manipulation of its function in crops and correction of its dysfunction in disease.
Defining the Brassica pan-genome and establishing methods for gene conversion based crop improvement. Gene content varies between individual varieties. Gene content varies between individual varieties. The project aims to apply novel genomic tools to identify and characterise the fixed and variable gene content in the important crop canola and use this to understand genome evolution as well as develop tools to accelerate canola breeding. The project team have developed and used a high-resolution genotyping approach to demonstrate that gene conversions, short recombination events which lead to the non-reciprocal exchange of genomic regions during meiosis, are abundant in crop genomes. The project aims to develop methods and resources to characterise gene conversion in canola and establish a basis for gene conversion based crop improvement.Read moreRead less
Developmental regulation of plant mitochondrial genome structure and copy number. Recombination is a major driving force behind mitochondrial DNA evolution and is responsible for the occurrence of cytoplasmic male sterile plants that are used by plant breeders to obtain high yield hybrids. A better understanding of the mechanisms that underlie mitochondrial and chloroplast genome maintenance and segregation will be a major fundamental scientific advance that will permit an integrated picture of the interactions between the three plant genomes (nuclear, mitochondrial and chloroplastic). It is also a pre-requisite for the future manipulation of the cytoplasmic genomes leading to new ways to develop varieties with modified cytoplasms.Read moreRead less
Saving seagrass from climate change. This research aims to test whether seagrass ecosystems can be safeguarded from climate change impacts by enhancing genetic connectivity in range edge populations using novel genetic rescue approaches. We will use the range edge seagrass meadows of the UNESCO World Heritage Site of Shark Bay as our model, which was significantly impacted by a marine heat wave in 2010/2011. The project will generate new knowledge on how seagrasses can adapt and survive in situ. ....Saving seagrass from climate change. Expected outcomes are improved conservation, management and restoration practices for seagrass meadows. This should provide significant benefits for long-term resilience of this economically and culturally significant ecosystem.Read moreRead less
