Summary
Chemotherapy treatment and the natural ageing process leads to ovarian failure and infertility. For young cancer survivors, chemotherapy induced infertility is devastating, and like aged women, leads to hormonal changes that lead to osteoporosis, diabetes, obesity, and neurodegeneration. Here, a new class of molecules will be tested for their ability to prevent and reverse infertility folllowing chemotherapy treatment, and in old age. These findings will improve the healthy lifespan of women.

Summary
Ovarian cancer is the 6th most common cause of cancer death in women and the proportion of women who die from their disease has not improved substantially over time. This large-scale study will use de-identified data from the Pharmaceutical Benefits Scheme, the Australian Cancer Database and the National Death Index to investigate whether medications commonly used for other conditions can help decrease the risk of ovarian cancer developing or improve survival from ovarian cancer after diagnosis.

Observational studies suggest that modifiable risk factors such as low vitamin D levels, coffee consumption, alcohol consumption and obesity may be important in cancer risk. However, observational studies can only demonstrate association between a risk factor and cancer, and association does not equal causation. We present an alternative approach to help determine which risk factors actually cause cancer.

Summary
The most lethal type of ovarian cancer, high-grade serous cancer (HGSC), can be divided into four subtypes based on gene patterns. One subtype involves a set of genes/proteins that, in their specific combination, result in activation of a pathway known as MYCN. As most HGSC start in the fallopian tube, we are using fallopian tube material to make new MYCN HGSC models to observe development in the earliest stages. We hope to generate new tests and treatments for this subtype of ovarian cancer.

Summary
Women diagnosed with ovarian cancer typically undergo surgery, followed by chemotherapy. However, the efficacy of chemotherapy varies widely, with some women responding well, whilst others are exposed to the toxic effects of a treatment that does them little good. We aim to identify the genes which explain why there are differences in response. This will lead to more individualised chemotherapy and improved outcomes for women with ovarian cancer.

Infertility is common and although IVF is widely accepted, the procedure is expensive and is associated with health risks. Using laboratory animals, we have made significant advances towards developing new technologies that can mature eggs and produce embryos in vitro, but without women receiving hormone injections. This project will seek means to combine the benefits of two of our existing technologies into one integrated system, to provide hormone-free infertility treatment.

Mucinous ovarian cancer (MOC) is different from other ovarian cancers but few studies have characterized the genetic changes specific to this subtype. It is often confused with metastases from other organs and does not respond well to standard ovarian cancer therapies. If MOC is more similar to mucinous cancers from other organs than other ovarian cancers, it may be better treated with chemotherapeutics that show success with other mucinous tumours.

Ovarian cancer is a major cause of cancer death in women because current treatments are inadequate. Half of aggressive ovarian cancers have abnormalities in DNA repair and should be susceptible to new PARP inhibitor therapy, yet not all those respond. By developing a new model of studying human ovarian cancers in mice, we can discover markers to predict which ovarian cancers will respond best to these exciting new treatments.
