Summary
Bowel cancer is a major health burden. Surgical resection of the primary cancer is often possible at diagnosis, yet in many patients the cancer will recur. Together with mutations, inherited genetic variants influence the rate of bowel cancer growth. This study aims to identify inherited variants predictive of cancer recurrence. Improved prediction of recurrence will permit more targeted use of clinical interventions, tailored to the individual patient, ultimately improving patient survival.
The recent cloning of the breast cancer predisposition gene, BRCA1 had an unexpected consequence. Whereas mutations were found in affected individuals from families showing a predisposition to breast and ovarian cancer, mutations were not identified in breast cancers from individuals with no family history. We have identified a type of change called an epigenetic change affecting BRCA1 in breast and ovarian cancers. The Transcription Factor ZNF652: Deciphering Its Role In Breast Cancer.

Summary
From our continuing research we have shown a protein called ZNF652 is involved in cancer. This proposal focuses on the role of this gene in the spread of cancer within the breast and to other organs. We will determine its role in a process where a cancer cell changes its characteristics to make it more likely to spread to other tissues. Our preliminary results suggest ZNF652 could be a marker that will predict poor prognosis. This proposal will further investigate this finding.

Breast cancer is the most malignant tumour of women and, despite great advances in detection and treatment, some 30% of women who present with primary breast cancer eventually relapse or die of their disease. Genetic studies have resulted in the rapid identification of the one-third of women at high risk of developing breast cancer because of a family history of the disease: it is hoped that these women will eventually benefit from advances in gene therapy now being developed. Genetic studies have resulted in the rapid identification of the one-third of women at high risk of developing breast cancer because of a family history of the disease: it is hoped that these women will eventually benefit from advances in gene therapy now being developed. For the majority of women developing breast cancer, however, the outcome, or prognosis, remains uncertain. The most important indicators of outcome are obtained by study of the excised cancer tissue, and these relate to the speed of growth of the cancer cells and their ability to migrate, or metastasise, to other sites in the body. Studies of cancer tissue using molecular cell biological methods has enabled the identification of several markers that are proving useful as indicators of outcome, and further understanding of the biological functioning of these markers will enable these molecules to be targetted in new treatments aimed at preventing the spread of the cancer. The present study will examine the appearance of new markers for cell migration among breast cancers and measure their value as indicators of outcome. One molecule in particular may be useful as a therapeutic target since it is used by migrating cells during development but is not expressed by normal (non-cancer) adult tissue cells. While most leukemia patients initially respond well to chemotherapy, >60% die because the disease returns as a result of the survival of leukaemia cells following treatment. We have identified a new protein, osteopontin (OPN), that may allow the survival of leukaemia cells and therefore reduce the ability of chemotherapy to erradicate disease. We seek to examine the role of OPN in leukemia with a view toward developing targetted therapies in the future.
