The interface between the cell and its environment is the cell membrane. Signals, nutrients, and ions all have to cross this barrier. In addition, the cells contain many specialized intracellular membranous compartments. We know little about the signals that direct the synthesis of these structures and determine their final composition and shape. This grant will utilize acid secretory cells in the stomach to examine these questions because they contain a very extensive membrane system. We will use a state-of-the-art genetic and cell biological technologies to manipulate and analyse these cells in a whole animal setting.Read moreRead less
Proteins form up to 25% of our diet. The first step in protein absorption is the digestion of protein into smaller units called peptides and amino acids. Both components are subsequently taken up by specialised cells in the wall of the intestine. In this project we plan to study how protein absorption occurs at the surface of these intestinal cells and investigate why this process fails in malabsorption syndromes, where the uptake of amino acids is impaired.
This proposal concerns the biochemical investigation of a protein called GABA receptor which is a known target for various anti-anxiety drugs (anxiolytics), anti-convulsants, sedatives, depressants, anti-epilespy drugs, alcohol and anaesthetics. This work is expected to lead to the determination of the three-dimensional shape of the protein which will provide essential information about how the protein works and lay the foundation for the design and development of new drugs to control epilepsy, ....This proposal concerns the biochemical investigation of a protein called GABA receptor which is a known target for various anti-anxiety drugs (anxiolytics), anti-convulsants, sedatives, depressants, anti-epilespy drugs, alcohol and anaesthetics. Molecular analysis of photosynthetically-linked, active CO2 uptake and CO2 signal transduction by cyanobacteria (blue-green algae). Cyanobacteria (blue-green algae) have evolved a very efficient means of capturing and concentrating CO2 for photosynthetic fixation into sugars, the basic building blocks for cell growth. This process is dependent on the operation of several unique, active uptake systems for CO2 and HCO3-, with their genetic expression regulated by CO2 supply. This proposal will capitalize on our progress in describing the functional genetics of this process and aims to elucidate the mechanism of active CO2 uptake and the way that cells sense the ambient CO2 concentration. The information gained is likely to be useful for designing improved crops.Read moreRead less
The role of mechanosensitive (MS) ion channels in magnetoreception. The magnetic field of the Earth has for long been known to influence the behaviour and orientation of a variety of organisms. The magnetic field of the Earth has for long been known to influence the behaviour and orientation of a variety of organisms. Experimental study of the magnetic sense has however, been impaired by the lack of a plausible cellular and/or molecular mechanism providing meaningful explanation for detection of magnetic fields by living organisms. Recently, mechanosensitive (MS) ion channels have been implied to play a role in magnetoreception. Based on our preliminary investigations, which suggest that the activity of bacterial MS channels may be affected by magnetic fields, we propose to study effects of magnetic fields on MS ion channels in Gram-negative bacteria Escherichia coli and Magnetospirillum magnetotacticum. The project promises also to contribute towards better understanding of adverse effects of electromagnetic radiation on human health and towards understanding the mechanisms behind remote magnetic-nanoparticle mediated activation of MS ion channels.Read moreRead less

The aim of this research, to understand the molecular organization and dynamics of the plasma membrane that underlie the signal transduction events, is at the very heart of understanding cell communication. T cell recognition and activation initiates an adaptive immune response to invading pathogens and structurally altered proteins that can be found in cancers. By providing functional insights into the molecular mechanism of T cell activation, we will not only provide fundamental knowledge of receptor signalling but also specific details of T cell receptort triggering that may lead to the development of new therapeutic strategies to control T cell activation.Read moreRead less

The secretory pathway of eukaryotic cells is fundamental for proper cell growth. The Golgi apparatus is a key organelle of this pathway where newly made proteins are selectively packaged into membrane-bound transport vehicles and then shipped to their correct destination, such as the surface of the cell. This research aims to understand the mechanism by which these cargo-loaded transport vehicles are generated from the Golgi apparatus. This information is of fundamental importance in understanding how a cell survives and grows, and is necessary to allow a rational basis for the engineering of secreted recombinant molecules.Read moreRead less

Using computer-based searches of genome sequence data, we now have a complete list of tail-anchored membrane proteins in the yeast Saccharomyces cerevisiae. These include a number of essential proteins, such as SNAREs and TOM proteins responsible for building cellular membranes in all organisms, including man. Of the additional protein sequences discovered in the search, 8 represent proteins of known function while 19 are novel. We propose to study the subcellular location of these 19 novel proteins, and solve how they are targeted to and inserted in membranes. We will also investigate the function of the newly-discovered proteins.Read moreRead less
