Understanding the immune response is proving extremely complex and promising results for disease treatments from animal models are often difficult to translate to new clinical therapies. My research is unearthing weaknesses in our current knowledge of the immune system and seeking to replace them with a foundation that can exploit new developments in computer modelling and systems biology. In this way I aim to rationally manipulate the immune response.

I am a molecular-cell biologist investigating the genetic control of lymphocyte differentiation. I study the mechanisms of action of transcription factors expressed specifically in B cells that regulate B lymphocyte specialisation and function, that determine the ability of mature B cells to respond to signals from other cells or from invading pathogens, and that enable the differentiation of antibody-secreting cells, the effectors of the B cell lineage. This application seeks information on the factors controlling T cell survival, tolerance and responsiveness to foreign antigens and tumour antigens. Particular attention will be directed to determining how T cells are kept alive through contact with self ligands and cytokines while preserving self tolerance and how anti-tumour responses can improved without augmenting the function of T regulatory cells.
Regulation Of Leukocyte Recruitment In Inflammation
Funder
National Health and Medical Research Council
Funding Amount
$686,656.00
Summary
In inflammatory diseases such as asthma, arthritis and atherosclerosis, white blood cells enter affected tissues causing inflammation and tissue destruction. This research will investigate the processes whereby white blood cells enter affected tissues, particularly how they exit the circulation, and migrate throughout tissues during inflammatory responses. An improved understanding of this process may identify new ways of interfering with the disease process in various inflammatory diseases.
The human immune system comprises many different types of cells that can detect foreign molecules. My research will lead the way to understanding some of the most abundant, yet least well understood, cells within this system, collectively known as 'unconventional T cells'. This knowledge is essential to optimally and efficiently manipulate the immune system in health and disease.
Defining The Coordination Of Immune Responses To Pathogens
Funder
National Health and Medical Research Council
Funding Amount
$640,210.00
Summary
Understanding how immune responses are coordinated is critical for the design of new therapies and vaccines to target infectious diseases and cancers. This project will utilise advanced imaging combined with novel tools to dissect the complex interactions that occur between immune cells as they are activated and patrol the body to eliminate infectious pathogens.
Transcriptional And Metabolic Regulation Of Effector And Memory Lymphocyte Differentiation

Funding Amount
$707,370.00
Summary
I am an internationally recognized expert in the field of lymphocyte biology. My work has shed light on antibody production, T cell responses and immune pathology. Specifically, I have identified molecular regulators that link antigen recognition, lymphocyte population expansion, cellular metabolism and effector function. My ongoing work focusses on the development and function of several critically important cell types, including tissue resident lymphocytes and regulatory T cells.
Mapping The Molecular Blueprint For Immune Cell Differentitation

Summary
Killer T cells are white blood cells that are key for helping control virus infections and in the recognition and elimination of cells that have become cancerous. This proposal aims to identify novel molecular mechanisms that control the ability of killer T cells to mediate their antiviral and anti-cancer functions. This will provide molecular targets for possible clinical interventions designed to either promote immunity (vaccination) or limit damage caused by T cell responses that target self
Delineating Immune Circuits For Innate And Adaptive Immune Protection

Funding Amount
$876,005.00
Summary
The immune system provides the essential frame-work to protect us against infection, disease and to heal tissues after trauma. This is achieved by a complex but elegant network of different types of white blood cells. Understanding the molecular wiring of these cells will provides fundamental insights to how the body fights pathogen infections and cancer and lays the foundation to therapeutic approaches to vaccination and disease treatments.
