Summary

This project is designed to understand to how Type-2 NKT cells develop within the body, how they recognise lipid molecules and how they carry out their immune functions. This work will have important implications in understanding the role of NKT cells in human health and disease.

Summary
Obesity increases the risk of miscarriage during pregnancy. The reasons for this are not known, although it is thought that abnormal levels of hormones and metabolic parameters are a contributing factor. We hypothesise that the immune system plays a role. In this project we will determine if obesity upsets the fine-tuning of the immune system that is crucial for successful pregnancy. Understanding the reason behind adverse pregnancy outcome will allow appropriate management of maternal obesity.
NKT cells are a type of lymphocyte with the ability to prevent a broad range of diseases including cancer, autoimmunity, infection and graft rejection. Sometimes, NKT cells also cause diseases, including allergy and atherosclerosis. This project will investigate how NKT cells become activated in these diseases by studying their surface receptors and what these receptors interact with.

Summary
Protection against infection depends on T cells, which recognise and target foreign molecules called antigens via a specialized antigen receptor known as a T cell receptor (TCR). This project focuses on a specialised population of T cells called Natural Killer T (NKT) cells that recognise lipid antigens. The work outlined in this grant will increase our understanding of how different TCRs can recognise diverse lipid antigens thus providing insight into the role of NKT cells in the immune system.
Prof Carbone’s laboratory specialises in the study of immunity at the body surfaces. These surfaces include skin and mucosal tissues such as respiratory and gastrointestinal tract, all of which are the common points of entry for a variety of infectious agents. He has found that there exist cells at these peripheral sites that are separate from the immune components that one finds in the blood and, more importantly, that provide a profound level of protection during new infection. Immune responses require the coordinated interaction and cross-talk between two types of white blood cells known as CD4 and CD8 T cells. A dysregulated interaction between these cells could be the cause of autoimmune and persistent infections by pathogens leading to chronic diseases.
