Summary
Thalassaemias are the most common hereditary diseases effecting the production of red blood cells. The underlying cause of disease is a failure to produce normal quantities of haemoglobin (Hb; the essential oxygen-carrying molecule in blood), resulting in severe anaemia. We have discovered a new protein with an important role in Hb production. We will elucidate the function of this protein in red blood cells and investigate novel treatments for thalssaemia disease.

Summary
In breast cancer the spread of cancer (metastasis) to bone occurs frequently and causes significant problems including pain, fracture, immobility and paralysis. We have recently discovered that a drug, widely used in Japan and Korea for skin disorders, inhibits breast cancer growth in bone using animal model systems of this disease. This is a very exciting and novel finding. We will further investigate the potential of this drug and identify precisely how it works at the molecular level.

New toxins including omega-conotoxin CVID have been discovered from fish eating cone snails found on the Great Barrier Reef. These toxins discriminate between ion channel receptors on nerve cells that allow calcium ions to enter the cell and nerve transmission to continue. Of great importance is the fact that CVID specifically and potently blocks nerves that transmit pain responses. A nucleosome is capable of blocking gene expression therefore one important function of chromatin is to prevent unwanted gene expression which is essential to allow an organism to develop properly. When gene expression is not accurately controlled by chromatin developmental defects or cancer could result from the production of incorrect proteins. To control correct gene expression, highly specific mechanisms must operate in the cell to remove, or disrupt, nucleosomes at certain genes at a precise time during development. One mechanism that we believe to be important is changing the make-up of a nucleosome. This can be achieved in the cell by the replacement of histones with different specialised forms of these histones (variants). It is thought that these histone variants could specifically expose certain genes and thereby turn them on. Once the correct protein is made in sufficient amounts the histone variants could be rapidly exchanged for the normal histones to shut off the gene. Employing a new approach, we will study one of these histone variants to discover the role it plays in turning genes on at precise times in early development during the formation of different specialised cell types. Recently we have discoverd new members of a family of growth fcators called vascular endothelial growth factors, and demonstrated their ability to promote the growth of blood and lymphatic vessels. In this study we set out to examine the role of another family of growth factor receptors, called RYK (for which we have a granted patent in the USA and Australia) in angiogenesis. functional experiments in mice have demonstrated that RYK can associated with a family of receptors called Eph receptors which play a key role in the remodelling of blood vessels during development and injury. Studying these molecules may tell us why blood vessels know' to be in the correct locations in the body and why in certain disease we see vessels of incorrect structure or location. These studies will form a basis of knowledge to develop rational means to manipulate blood vessel formation in the body, using non-surgical methods.
