A bird's eye view of sex: the chicken embryo as a model for gonadal development. The development of an embryo as either male or female (sex determination) is a fundamental biological process that fascinates both scientific and lay communities alike. This project will use the chicken embryo as a unique model organism to study how genes control sex determination. We will bring a novel method to our field of study; the use of avian viruses to deliver genes into embryos. This project will use the chicken embryo as a unique model organism to study how genes control sex determination. We will bring a novel method to our field of study; the use of avian viruses to deliver genes into embryos. This project will enhance our understanding of a basic biological process. It will have application to the poultry industry, in terms of sex ratio manipulation. It will also aid in the diagnosis of humans born with sexual abnormalities. The project will consolidate Australia's standing as a world leader in the area of reproduction and development.Read moreRead less

Calcium movements into and out of the cytoplasm of smooth muscle cells are regulated primarily by a variety of proteins located in the plasma membrane and in the sarcoplasmic reticulum and play a central role in controlling the contractile state of smooth muscle. Understanding the mechanisms that control intracellular calcium levels is fundamental to understanding smooth muscle function. This project will employ a unique approach, involving the use of mice with targeted disruptions to genes encoding key calcium transport proteins, to gain new knowledge on the contribution of various calcium handling pathways to overall control of smooth muscle function.Read moreRead less

The potential of the information gained from the lung imaging for improving the diagnosis and monitoring of adult respiratory diseases, such as emphysema, pulmonary fibrosis and asthma, is enormous. Abnormalities in the structure and function of the kidney circulation are likely key factors causing hypertension, so a detailed understanding of the kidney circulation is required before we can cure or prevent hypertension. The research underpinning cardiovascular and renal flows will assist in the understanding of hypertension, a major risk factor for cardiovascular disease, which accounts for approximately 40% of all deaths in Australia and particularly high for dialysis patients. This project will benefit the community by informing service provision for infertility treatment, and by identifying the potential for prevention strategies. It will consolidate Australia's position at the forefront of research into assisted reproductive technologies, through the extension of this expertise to our near neighbours. It will contribute to the current gap in knowledge of infertility in Southeast Asia and in developing countries more broadly, and advance research on infertility conducted from human rights and anthropological perspectives. The project will reinforce the relevance of the United Nations definition of reproductive health, for all people regardless of whether they live in highly or lesser developed nations.Read moreRead less

In Australia, about 30% of adults have hypertension, a major risk factor for heart disease, accounting for about 40% of all deaths. Problems in renal circulation are likely factors leading to hypertension. A detailed understanding of the renal circulation, of whose hydraulic characteristics we have limited knowledge, is required before we can cure or prevent hypertension. We will determine how the size, structure and geometry of the blood vessels influence the function of the kidney. This will lead to predictive models to aid the design and interpretation of physiological studies and the combat of hypertension. It will also help in the ongoing development of bioartificial kidneys to replace present dialysis systems. Read moreRead less

This project aims to develop a novel electrode as an integral component of a new treatment for severe stress urinary incontinence. Treatments for severe stress urinary incontinence are associated with complications and are not completely effective. The new electrode is designed to be activated by an implanted stimulator to control an innervated smooth muscle graft (the neosphincter) to regulate the flow of urine from the bladder. Project research into the design of the electrode will focus on providing safe, effective and efficient stimulation of the neosphincter, while ensuring minimal damage to the surrounding tissues and affording straightforward implantation at surgery.Read moreRead less
Development of an electrode assembly for the stimulation of a transplanted innervated smooth muscle sphincter. The project will identify an optimal electrode design, which will be used in future clinical trials of the device.
The role of SGK-1 and SGK-2 in hypertension and nephropathy in diabetes mellitus. The key objective is to define the suitability of the serum glucocorticoid regulated kinases -1 and -2 (SGK-1, -2) as novel drug discovery targets. The key objective is to define the suitability of the serum glucocorticoid regulated kinases -1 and -2 (SGK-1, -2) as novel drug discovery targets. A specific inhibitor targeting SGK-1 and -2 will be tested to determine if it reverses the enhanced sodium reabsorption and extracellular matrix production characteristic of progressive renal failure in in vitro models models of renal disease. These inhibitors present an opportunity to control hypertension whilst simultaneously limiting fibrosis in the kidney. Renal failure is steadily increasing and is now the single largest health care cost to the community. These studies will provide the proof of concept required to ultimately bring this novel preventative therapy to the community.Read moreRead less
