A bird's eye view of sex: the chicken embryo as a model for gonadal development. The development of an embryo as either male or female (sex determination) is a fundamental biological process that fascinates both scientific and lay communities alike. This project will use the chicken embryo as a unique model organism to study how genes control sex determination. We will bring a novel method to our field of study; the use of avian viruses to deliver genes into embryos. This project will enhance our understanding of a basic biological process. It will have application to the poultry industry, in terms of sex ratio manipulation. It will also aid in the diagnosis of humans born with sexual abnormalities. The project will consolidate Australia's standing as a world leader in the area of reproduction and development.Read moreRead less

We are studying a mouse model of abnormal sex organ development in which genetically female mice develop as males. Our basic research program will lead to greater understanding of the genetic switch controlling the formation of male and female characteristics. This research should in turn provide insight into the causes of defects in patients with disorders of sex development, helping to inform the difficult clinical decisions that need to be made for their treatment, and ultimately leading to better management and therapeutic strategies. Our studies may also provide unique methods to control the exotic mouse population, using the daughterless strategy.Read moreRead less

This project seeks to better understand regulatory mechanisms of embryonic development. A key challenge in biology is to understand how genes act in regulatory networks to coordinate the formation of complex organs during embryo development. Although several genes influence embryo sex determination and gonad development, Sox9 has emerged as a master regulator in this system. This project plans to use new systems biology technologies and transgenic mice to identify the suite of Sox9 target genes and partner proteins. Anticipated outcomes include insights into sex determination, stem cell differentiation, and development of many other organ systems that depend on Sox9 function, providing proof-of-concept for high-throughput analysis of molecular control networks in the embryo.Read moreRead less
Defining a role for non-coding RNAs in gonadal sex differentiation. This project aims to increase knowledge in the area of developmental biology, studying how gene regulation by so-called non-coding RNAs contributes to tissue patterning. The project plans to use a unique model system: gonadal development in the chicken embryo. It also plans to use novel molecular approaches that exploit the chicken model to study the role of microRNAs and a long non-coding RNA in patterning the embryonic gonad. ....Defining a role for non-coding RNAs in gonadal sex differentiation. The project plans to use a unique model system: gonadal development in the chicken embryo. It also plans to use novel molecular approaches that exploit the chicken model to study the role of microRNAs and a long non-coding RNA in patterning the embryonic gonad. The project aims to provide a deeper understanding of how genes operate to control tissue patterning and organogenesis. It may thus inform the field of sex determination specifically, and, more broadly, stem cell biology and tissue engineering.Read moreRead less
The structure and patterning of branching morphogenesis in the developing kidney. This project aims to understand a fundamental developmental process known as branching morphogenesis, which drives the formation of many organs including the kidney, lungs and glands. Understanding this process will be of key importance in understand how our organs form.
Morphological development of the kidney – a paradigm for organogenesis. This project aims to shed new light on how the kidney develops, how normal adult function is established and how congenital kidney malformations occur. It aims to investigate a mathematical pattern that has been discovered in the program of branching morphogenesis which dictates kidney development. By combining cutting-edge genetic approaches, morphological analysis and mathematical modelling, this project plans to explore h ....Morphological development of the kidney – a paradigm for organogenesis. This project aims to shed new light on how the kidney develops, how normal adult function is established and how congenital kidney malformations occur. It aims to investigate a mathematical pattern that has been discovered in the program of branching morphogenesis which dictates kidney development. By combining cutting-edge genetic approaches, morphological analysis and mathematical modelling, this project plans to explore how this pattern is established, how it dictates kidney development and whether it constitutes an ancient mechanism central to organ formation. Project outcomes may improve understanding of fundamental developmental processes and developmental disease and variation. Project findings may also be important for understanding the development of other organ systems.Read moreRead less
Switching on sex: How key mammalian sex-determining genes are activated. Development as a male or female is one of the most important biological decisions made during embryonic life. Development as a male or female is one of the most important biological decisions made during embryonic life. Although several of the key sex-determining genes are now known, it is unclear how they come to be active at the right time and in the right cells of the gonad to direct sex determination. This project aims to use bioinformatics, biochemistry, cellular assays and powerful genome editing technologies in mice to pinpoint essential regulatory elements and understand how they function. In addition to addressing key issues in mammalian molecular genetics and developmental biology, data generated in this project may provide a basis for innovative biotechnologies in pest management, wildlife conservation and livestock breeding.Read moreRead less
