Viruses that cause serious diseases such as hemorrhagic fever or encephalitis must be quickly identified. Diagnostic tests based on DNA hybridisation are accurate and can be rapid but they are expensive. We will test a method for simplifying DNA tests and increasing their capabilities. DNA probes for detecting arboviruses will be designed at the ANU using new bioinformatic methods and their reliability will be modelled using all the available genetic information. Computer predictions will be experimentally tested in the PANBIO laboratory by using the probes to detect viral nucleic acids. The influence of virus genome complexity will be investigatedRead moreRead less

West Nile virus (WNV) is transmitted by mosquitoes and causes a fatal disease of the brain in animals and humans. WNV is currently exotic to this country, however a closely related, but benign cousin of this virus (Kunjin) can be found in Northern Australia. Currently it is very difficult to differentiate between infections with WNV and Kunjin using standard laboratory tests, therefore it will be difficult for public health officials to detect WNV if it is introduced to Australia. In this project we aim to develop more accurate laboratory tests to detect infections with the deadly, exotic WNV using new strategies. Read moreRead less

Indigenous health and welfare is of major concern for Australians. This project, which focuses on the dog, also recognises that the dog harbours a number of diseases that can infect humans. To maintain sustainable dog health programs to reduce disease in dogs, and, as a corollary, have positive impact on human health and welfare, culturally-relevant, evidence-based education programs are critical. Dog health programs will indirectly improve the expectations, standards and self-worth of many Indigenous Australians. As a consequence, the national benefits include the development of environmentally sustainable Indigenous communities, and the strengthening of Australia's social and economic fabric especially in rural and remote areas.

Cryptosporidium is the major public health concern of water utilities as the parasite has a global impact on the health and survival of millions of people and animals worldwide. It is resistant to chlorine and there are no effective drugs against it. Control strategies therefore rely on understanding how it is transmitted. This project will conduct a comprehensive study utilising molecular tools to determine if the Cryptosporidium in humans is the same as that derived from water contamination sources (animals, sewage etc). The outcomes of this project will result in the development of more targeted, cost-effective preventive measures to minimize exposures to infections, accurate risk assessment, and scientific management of the watershed.Read moreRead less
Development of an ultrasensitive assay for human prion proteins. The aim of this work is to enable the detection of prion proteins in human blood and other tissues. The outcome of the work is expected to facilitate the production and certification of prion-free blood and blood products. Prions cause Bovine Spongiform Encaphalopathy and Creutzfeld Jacob Disease and attempts to control of these diseases would be greatly abetted by an optimal test for the disease-causing agent.Read moreRead less

The project aims to use the technique of real-time polymerase chain reaction to rapidly detect and quantify the organisms associated with emerging and re-emerging infectious diseases of man and animals. It will also be used to determine related gene expression.
The equipment will be used to support a wide range of projects that require the detection of specific RNA or DNA and it will allow the rapid, cost effective and efficient processing of either RNA or DNA from large numbers of samples. Minor variations in organisms will be detected using this equipment.

Genetics and evolution of Shigella O antigens. We use genome scale sequencing techniques to sequence 26 O-antigen gene clusters from Shigella. With the seven already known, this will give sequences for every O-antigen of Shigella. This will be the first time that such set is fully sequenced. Shigella are human specific pathogens, have emerged with the evolution of humans. O-antigens are important for their life and pathogenicity. This will be the first time that such set is fully sequenced. Shigella are human specific pathogens, have emerged with the evolution of humans. O-antigens are important for their life and pathogenicity. This project will greatly extend our knowledge of the genetic basis and evolution of this important polymorphism. O-antigens are used for typing Shigella and also elicit strong immunity. The molecular data will help establish DNA based typing and vaccine development.Read moreRead less
Functional Genomics to Predict and Enhance Response to Interferon. The increasing number and huge cost impost of new therapies to health providers, both worldwide and nationally, has not yet resulted in a concomitant increase in strategies to optimise their use. Many of the new therapies are proteins (recombinant human proteins or humanised monoclonal antibodies). The improved use of one of Australia's most expensive commonly used protein drugs, pegylated interferon ribavirin (Peg-IFN-R), could potentially produce savings to the Pharmaceutical Benefits Scheme (PBS), and improve delivery of healthcare to thousands of Australians.Read moreRead less
The biosynthesis of structural proteins in parasites. The socio-economic impact of parasitic diseases, in Australia and throughout the world, is enormous- they kill 2-3 million people per year and cost livestock industries billions of dollars per annum. Some are serious food and waterborne threats, such as the coccidia, because they are transmitted from person-to-person via the faecal-oral route, or via ingestion of contaminated water or food, or through the ingestion of cysts in raw or undercooked meat. They cause diarrhoea, which in some cases, can be life-threatening. We will understand how the coccidia protect themselves as they move from host to host and, through that understanding, develop new ways to control them and eliminate the suffering caused by parasitic diseases.Read moreRead less

This project aims to generate increased understanding of the aetiology and pathogenesis of the two most prevalent and debilitating infectious diseases of koalas to make improved decisions regarding therapeutic intervention, prevention and management. Chlamydiosis, caused by two chlamydial bacteria, affects koala fertility, urinary tract function and eyesight. Cryptococcosis, caused by the fungus Cryptococcus gattii, may result in koala morbidity and death. Planned therapeutic trials, prevention strategies and the development of optimal husbandry strategies will provide knowledge that will lead to reduced koala morbidity through improved understanding of the interactions amongst infectious pathogens, the animal host and the environment.Read moreRead less
