Summary
Papillomavirus (PV) is a sexually-transmitted virus that is a major cause of cervical cancer. Our study will determine how PV is able to form new virus particles inside infected cells. This is a critical part of the virus life-cycle, and a better understanding of this process may allow it to be trageted by anti-viral treatments. In addition, we will develop a method to create non-harmful virus particles which we will use to study human immune responses to the virus.

Understanding why, when, how and at what pace the HIV virus hides and establishes itself in one's body will allow us to design new ways for preventing and eliminating this reservoir of hidden HIV. As a clinician scientist in HIV and infectious diseases, I will drive clinical studies to explore the kinetics of HIV in patients who recently acquired HIV, those who start HIV treatment early, and those chronically infected with HIV so as to investigate novel means to minimise HIV hiding spots.

Summary
Despite the eradication of smallpox by vaccination, poxviruses remain a threat to public health because of bioterrorist scares from kept variola stocks and because of the possible emergence of other poxvirus pathogens from the extensive animal reservoir. The structural analysis of the assembly of poxvirus will not only improve our knowledge of fundamental processes, highly conserved in DNA viruses, but could also provide valuable targets for the rational design of antiviral drugs.

Australia is currently in the process of manufacturing vaccines for use in people against strains of avian influenza viruses circulating in South East Asia as part of a national preparedness program for an influenza pandemic. These particular avian flu viruses are capable of causing severe disease and death in humans as well as birds, although at present they are not highly transmissible between people. Should the avian influenza viruses mutate to gain this capability, it will be necessary to institute widespread vaccination of the Australian population. It is not possible to test the vaccines in people for their effectiveness against avian influenza infection prior to a disease outbreak, so an animal model for the disease will be used to assist in optimizing the formulation of flu vaccines and in testing their efficacy in preventing infection or reducing the severity of disease. Ferrets are natural hosts for flu viruses, have similar responses to vaccination as people, and develop a similar disease to humans when infected with influenza. Despite the great successes in antiretroviral therapy (ART) in reducing HIV-associated mortality, treatment is life long and there is no cure. The major barrier to a cure for HIV is the persistence of long lived latently infected cells on ART. Over the next five years I will discover, develop, optimise and evaluate novel interventions to eliminate latently infected cells, long lived infected cells in the liver and enhance HIV-specific immunity through immunotherapy.
Development and commercialization of novel diagnostic assays for the early detection of acute dengue virus infection. Dengue is an emerging disease of the tropics and is endemic in more than 100 countries with up to 100 million cases annually. Of these, 500,000 result in dengue haemorrhagic fever (DHF), a serious life-threatening complication of dengue virus infection.
