Summary
Novel Optical Microprobes - fluorescence excitation, lifetime and surface enhanced Raman spectroscopies. State-of-the-art technologies in microscopy will be combined with the versatility and diagnostic power of spectroscopy for the analysis and identification of materials by methods sensitive to chemical structure as well as spatial inhomogeneity. The design and synthesis of angiotensin converting enzyme-2 (ACE2) inhibitors. A vast number of current drugs on the market are inhibitors of enzymes whose action needs to be controlled in order to treat many conditions. This proposal will apply our new approaches to the design of enzyme inhibitors with superior therapeutic action. Metal catalysed reactions in organic synthesis. The major aim of this project is to develop highly efficient metal-catalysed routes to pharmaceuticals, e.g. new antibiotics and other commercially important organic molecules.
Particular attention will be paid to developing systems which facilitate the addition of readily available, cheap small molecules, e.g. hydrogen and carbon monoxide, to unsaturated organic molecules with high regio- and stereoselectivity.

The development of yoctowells on magnetic nanoparticles as both tiny chemical reactors and biological models. This project seeks to develop an innovative and cutting-edge research program in biomimicry by studying a surface functionalised system - the so called yoctowells - on magnetic nanoparticles, by studying their inclusion behaviour and utilising the intrinsic magnetic properties for isolation and manipulation in catalysis, medicine and electronics.
Development and application of new peptide ligation methods for the synthesis and structure-function studies of glycoproteins. Novel synthetic technologies will be developed in this project to facilitate the preparation of glycoproteins, which are of widespread biological and therapeutic interest. These methods will enable the preparation of pure glycoproteins for detailed biochemical and functional studies eventually leading to therapeutic and diagnostic applications.

This project aims to address a lack of knowledge about how post-translational sulfation of tyrosine residues influences protein function. The project will develop a synthetic platform for the rapid and efficient generation of libraries of site-selectively sulfated proteins. Enantiomeric characterisation allows evaluation of drug purity for potentially harmful constituents, consumer protection against adulteration of essential oils and natural materials, and study of chemical interactions in biochemical/environmental systems. Chromatograph technologies (GC, HPLC and CE)for enantioseparation are available, however limitations prevent multicomponent characterisation of complete sample mixtures. Critical choices involve: choosing a chiral selector that interacts differentially upon all enantiomers; incorporation of selector into high-efficiency chromatographic systems; application to all target compounds in the sample. We will synthesise novel chiral selectors, incorporate these into new separation columns, apply ultra-high resolution 2-dimensional GCxGC to broad-spectrum chiral analysis of volatile chemical samples.Read moreRead less
