Summary
Current treatments for age-related diseases of the central nervous system (CNS) are limited. We have shown in animal models of acute CNS degenerations that treatment with saffron or low energy infrared light is strongly protective. This project will determine if these treatments prevent CNS damage and dysfunction in animal models of chronic degenerations and add to knowledge of how these treatments work. This research should lay the foundation for testing these novel treatments in humans.
Vitamin E-bisphosphonate Conjugates: A New Therapeutic Approach Targeting Pathological Bone Loss Associated With Osteoporosis

Summary
Approximately 2 million Australians have osteoporosis, a disease which increases the risk of fracture and whose monetary and social costs are large and growing. The objective of this proposal is to exploit the osteotropic properties of novel compounds of vitamin E (VE) derivatives (BPVE) conjugated to bisphosphonates (BP), which will have anti-oxidant, anti-osteolytic and bone anabolic properties, and will be taken up selectively in the skeleton for the treatment of osteoporosis.
According to current National Heart Foundation surveys high blood pressure affects almost 3 million Australians, and accounts for about 6% of the total burden of disease. Unless effectively treated, high blood pressure may result in serious complications such as stroke, heart attacks and heart failure. The cause of high blood pressure in most people is unknown, but there is consistent information from population studies that low vitamin C intake is associated with high blood pressure. Recently, it has been shown that a diet rich in fruit and vegetables can lower blood pressure. We recently demonstrated in a small study that vitamin C tablets for one month lowered blood pressure, suggesting that vitamin C was the key beneficial element of the fruit and vegetable diet. Interestingly, some recent work suggests that the recommended daily allowances of vitamin C are less than a third of what they need to be to get effective levels of vitamin C in body tissues. Studies in animal models of high blood pressure have shown that oxidant levels are increased. This can affect the ability of blood vessels to relax, and may be a cause, rather than a result of high blood pressure. It seems possible that vitamin C (a strong antioxidant) may be having its beneficial effect by reducing 'oxidant stress' in blood and blood vessel walls. The purpose of this project is to further determine the effect of oxidants and antioxidants (specifically vitamin C) on blood pressure and blood vessel function in patients with high blood pressure. It is clearly prudent to eat a diet rich in fruit and vegetables, but it is important that we understand why this is so, and whether specific dietary components are critical. Our goal is to target natural reduction/oxidation (redox) modifiers, i.e. niacin and sulforaphane, to the skin deeper layers to treat photoageing. Both drugs have been separately shown to prevent UV induced skin cancer. The outcomes of this project will be safer and more effective prevention and treatment of sun damaged skin. This project can provide health benefits to Australians from improved treatment of sun damage, and economic benefits to the nation.

Friedreich ataxia, a life-shortening neurologic condition, has no proven treatment. This study will assess a medication called resveratrol in 60 individuals in centres in four Australian states. The second study will explore the effectiveness of an emerging imaging technique, high-resolution nerve ultrasound, in the diagnosis and monitoring of conditions that affect the nerves in childhood.

This project examines the potential for dietary carotenoids to be used to manage asthma. Carotenoids are antioxidants that are found in orange and red fruits and vegetables, such as tomatoes, carrots and mangoes. Research suggests that dietary carotenoids may be protective against asthma symptoms and -or onset. It is also likely that increasing intake of carotenoid-rich foods may be more effective than taking dietary supplements, as the key nutrients or combination of nutrients may not be known. This project will examine whether carotenoids such as lycopene can reduce the tendency of asthmatic airways to overreact to common triggers. It also investigates whether carotenoids can be used to prevent or reduce the severity of asthma attacks. The project will determine whether tomato juice or lycopene capsules are more effective in this role. This work will provide the necessary information to develop a large trial testing the ability of carotenoids to improve quality of life for people with asthma. While there is evidence to suggest that carotenoids may be helpful in asthma, the data to date is inconclusive. Intrauterine fetal growth restriction (IUGR) is a serious complication of pregnancy associated with increased perinatal morbidity and mortality. In particular, IUGR infants have a high risk of perinatal brain injury which is likely to arise from damage before birth. Our aim is to use an ovine model of IUGR to define the causes of that brain injury and to develop new therapies that could be offered to women to protect their unborn baby.
