Summary
Familial and young onset bowel and uterine cancer are usually caused by the inheritance of spelling mistakes in the genetic code within a set of cancer-protection genes. Recently, some patients were identified with their gene switched off by paralysing chemicals instead. This study aims to identify additional cancer cases with gene paralysis, determine if this arises in the presence or absence of a genetic change in front of the gene, and how gene paralysis is transmitted to the next generation.
The proposed program of work aims to shed light on dietary, environmental and genetic factors that may be related to the risk of childhood cancers. Identifying these risk factors, and how they interact with each other, will provide clues as to how childhood cancers may be prevented. The research program includes a number of Australian and international studies, which will involve collaboration between doctors and research scientists from a range of disciplines.

The treatment of diabetes involves the use of drugs that stimulate the release of insulin from the pancreas to reduce the high blood sugar levels. However, we believe that while in the short term this is a good strategy, in the long-term it damages the cells that produce insulin leading to a worsening state of diabetes. It is the aim of this application to understand the mechanisms by which the insulin producing cells are damaged when forced to oversecrete insulin.

Data from this study could potentially define new standards of clinical practice, as they relate to follow-up screening for response to therapy and recurrence of disease.

Kinase inhibitors are some of the most successful anti-cancer agents that have emerged in the last 15 years. However, tumors become resistant to these drugs after showing initial response. Understanding mechanisms through which cancer cells become resistant to these drugs will allow us to develop effective strategies to counter it and achieve sustained responses to cancer therapy. I propose to build a research program to systematically study these mechanisms to improve cancer therapeutics.
