Summary
Does melatonin work for children with ADHD and initial insomnia? We will study children with ADHD and initial insomnia using N-of-1 trials. This is a trial in a single person, which allows them the opportunity to learn if melatonin is effective for them.We will compare the usual clinical trials (RCTs, which do not give individual results) with combined results from all the 270 patients who have done N-of-1 trials. Data from the first part of each person’s n-of-1 trial will form the RCT.

Summary
In schizophrenia ability to maintaining focused attention is impaired. We explore how problems in “next state” prediction contribute to problems in attention. The brain constantly predicts what state of activation it will be in next. When events match these predictions we can easily ignore them but if predictions are wrong a prediction–error can trigger attention interruptions. We will test whether problems in prediction-errors make persons with schizophrenia more susceptible to distraction.

Understanding the conditions that confer vulnerability to the triggering of schizophrenia by cannabis use is key to preventing transition to psychosis in up to 14% of cases. This project will examine the long- and short-term effects of different cannabis compounds on specific vulnerability markers of brain chemical abnormalities in cannabis users from the general community.

Summary
Alzheimer’s disease is characterised by the deposition of amyloid plaques in the brain. We don’t fully understand how amyloid deposition occurs and what contribution is made by genetic and environmental factors. Amyloid deposition in the brain can now be quantified during life using positron emission tomography. In this study, we will examine brain amyloid in twins, which will determine what proportion of the pathology is attributable to environmental factors that may be modifiable.

The AHEAD Study will assess the impact of anaesthesia and surgery on cognitive function in patients with Mild Cognitive Impairment or mild Alzheimer's Disease. Such effects have not been established. With an ageing population being increasingly exposed to healthcare procedures requiring anaesthesia or sedation, any negative impact needs to be identified so that therapeutic decisions may be informed and future research appropriately targeted.

Summary
This proposal will provide essential new information on the role of deficits in decision-making associated with Parkinson’s disease dementia. We will use an innovative animal model to assess the influence of neurodegeneration and neuroinflammation the consequent loss of function in the neuronal systems supporting the learning and memory processes that contribute to goal-directed action, particularly the way new learning interacts with existing memory to guide choice and decision-making.
Attention deficit hyperactivity disorde(ADHD) is the most prevalent mental disorder of childhood affecting around 7.5% of Australian school age children. The disorder is strongly genetic and causes significant impairments in academic functioning, family and peer relations with sufferers at increased risk for drug abuse. Identification and characterisation of rare mutations will enhance our knowledge of the neurobiology and advance the search for next generation drug treatments for the disorder.

This study will evaluate the impact of a novel question prompt list (QPL) for parents of children with attention-deficit hyperactivity disorder (ADHD), on their communication with their child’s doctor, and health outcomes such as adherence. The QPL aims to increase parent question asking and information sharing between clinicians and parents; improve parents’ ADHD knowledge across the developmental lifespan of their child; and thus improve adherence to medication in children with ADHD.

Attention deficit hyperactivity disorder is a prevalent behavioural disorder affecting 7.4% of Australian children and adolescents. It has a strong genetic component with high heritability estimates (75–90%) comparable to other serious mental illness such as autism and schizophrenia. Attention deficit hyperactivity disorder (ADHD) affects 5% of Australian children. Up to 64% of children with ADHD experience impairing anxiety, which is associated with higher levels of ADHD symptoms and poorer quality of life. This randomised controlled trial will determine whether treating anxiety in children with ADHD improves outcomes.
