Summary
Many bacteria that cause diarrhoea act via the gut’s own nervous system to massively increase the transport of water and salt into the lumen of the intestine. In this project we will investigate mechanisms that lead to this over-excitation of the gut’s nervous system to test the idea that this is due to a change in the properties of the final nerve cells in the normal pathway that controls water transport. This will identify novel sites for drug treatments of diarrhoea.

Summary
In some people C. difficile bacteria naturally reside in the gut. Other people accidentally ingest spores of the bacteria while they are patients in a hospital or nursing home. Sometimes, broad-spectrum antibiotics used to treat an infection also kill healthy gut bacteria. The gut then becomes overrun with C. difficile, causing diarrhoea and pain, and sometimes death. We will investigate the use of a new potent antibiotic, vancapticin, to kill C. difficle and prevent relapse of infection.

Summary
Defects in the secretion of bile into the intestine cause digestive diseases, and abnormal circulating levels of bile acids induce profound itch and abnormal pain sensation. This project examines whether a cell-surface receptor (TGR5) produced by intestinal and sensory neurons mediates actions of bile acids on intestinal functions, itch and pain. The project will define mechanisms of digestive and sensory disorders and identify new therapies for constipation, diarrhoea, itch and pain.
