Summary
Omega-3 fats are a type of polyunsaturated fat which has long been thought to be beneficial for health. The body cannot make its own omega-3 fats - they must be taken in through the diet, or in supplement form. Large population-based studies have suggested that a diet rich in omega-3 fats can reduce the risk of cardiovascular disease, lower blood pressure, cut the risk of cancers such as prostate and skin. The fats have also been touted as a treatment for arthritis, asthma, and inflamed bowels. ....Omega-3 fats are a type of polyunsaturated fat which has long been thought to be beneficial for health. The fats have also been touted as a treatment for arthritis, asthma, and inflamed bowels. A lack of omega-3s during foetal development can hinder brain and eye development. Other work has suggested that a diet rich in omega-3 may help improve mental health. Studies revealed beneficial effects in a broad range of mental health problems including violent and aggressive behaviour in teenagers, major depression, bipolar affective disorder, borderline personality disorder, and schizophrenia. Evidence that omega-3 fatty acids may reduce symptoms in schizophrenia and other major psychiatric disorders make omega-3 fatty acids an optimal candidate for further evaluation of its therapeutic efficacy, in particular in young people in early illness stages of psychosis where specific intervention is controversial due to potential risks and medication side-effects. The proposed 12-week intervention study aims to establish a new treatment strategy alternative to antipsychotic medication in the young people with subthreshold symptoms at high risk for psychosis. Omega-3 fatty acids have no clinically relevant side effects which often accompany therapy with antipsychotics (such as diabetes and weight gain). A widely available, easy applicable, biological treatment without clinically relevant side effects could help to improve compliance and outcome in psychosis. Schizophrenia is often prolonged and results in a considerable economic burden. Age-related diseases are becoming a major concern as the world’s population grows older due to advances in medical technology, health and nutrition. Dementia accounts for a large proportion of agerelated diseases and is characterised clinically by deterioration in memory and cognitive processing. AD is the most common form of cerebral degeneration leading to dementia. Currently over 200,000 Australians suffer from dementia, with AD, accounting for 50-70% of all cases. At this rate, the number of people in Australia with dementia will increase to 730,000 by the year 2050. Thus, there is an urgent need for effective preventative treatments for this devastating disease, as dementia will soon be the major cause of disability in Australia. As a result, the social and economic consequences of this disease present a significant challenge to society, and it is imperative that strategies to prevent or delay the onset of AD are developed. If complimentary and alternative medicine (CAM) therapies could be developed to prevent or delay the onset for Alzheimer’s disease, the impact on disease burden could be substantial. However, these CAM therapies need to be critically evaluated for their mechanisms, efficacy and safety before human clinical trial are undertaken. The proposed research plan will evaluate the efficacy of the nutritional supplements of polyphenols (EGCG from green tea and curcumin), omega-3 essential fatty acids (docosahexaenoic acid) and lipoic acid to determine whether these treatments in combination offer preventative therapies for AD. The findings from the current study will provide important information concerning the effects of combination supplements in preventing cognitive deficits and AD pathology in a transgenic mouse model. Combination treatments may reduce cognitive deficits (memory and learning), oxidative stress and AD pathology, thereby providing an important insight into possible CAM preventative treatment strategies for AD. The development of effective preventative strategies for the treatment of AD is critical if we are to reduce the number of people that are expected to develop AD over the next 50 years, due to the rapidly aging population. Some vegetable oils such as canola, echium, and flaxseed oils have omega-3 fats. However, these are not the same as the omega-3s in fish oil. In general, vegetable omega-3s are known as short-chain omega-3s whereas those in fish are long-chain. When the vegetable oil omega-3s are consumed in the diet, the body must convert them to the fish-type, or long-chain, omega-3s in order to gain the health benefits. Currently, human metabolism is poor at doing this conversion. Our studies suggest that the amounts and blends of fats in most diets are well below optimal for conversion to long-chain omega-3s in the body. In particular, our studies show that there may be no point in simply adding more vegetable omega-3 fats to the diet. This project will examine the enzymes that convert the short-chain to the long-chain omega-3 fats. In particular, it will examine how the dietary fats interact with each other in detrimental ways to suppress activity of the conversion enzymes necessary for long-chain omega-3 formation. Evidence for the health benefits of fish-based omega-3 fats is very strong and it is an approach to health that most people can undertake on their own. Our lack of knowledge of the best way to use plant-based omega-3 fats is a large impediment for a more sustainable and broader based use of omega-3 fats. The results from this study will begin to fill in this knowledge gap. The results will be useful across a range of activities that are determinants of the kinds of fat that we eat. The effects are consistent and quite strong and appear to be unrelated to blood cholesterol levels. The effects are more likely due to the possibility that fish oil, which contains n-3 fats, can suppress abnormal heart rhythms which can cause 'sudden cardiac death'. This has been shown in animal studies, and recently we have shown that they can also suppress abnormal heart ....Consumption of fish and fish oil has been associated with protective effects for deaths from heart disease in many studies. The effects are consistent and quite strong and appear to be unrelated to blood cholesterol levels. The effects are more likely due to the possibility that fish oil, which contains n-3 fats, can suppress abnormal heart rhythms which can cause 'sudden cardiac death'. This has been shown in animal studies, and recently we have shown that they can also suppress abnormal heart rhythms in humans. Patients undergoing coronary bypass surgery are particularly susceptible to abnormal heart rhythms in the upper chambers of the heart in the days immediately following surgery. While these abnormal rhythms are rarely life threatening, they can result in increased post-surgical complications and a longer hospital stay with associated increased costs to the health system. In this study we will determine whether providing high dose fish oil for 3 weeks immediately preceding coronary bypass surgery, will result in a reduction in the proportion of patients experiencing post-surgical abnormal heart rhythms. We will also examine the effects of n-3 fats on the incidence of other complications following bypass surgery and also the length of hospital stay. In addition, we will gather data which will provide insight into the mechanisms by which n-3 fatty acids produce their well documented benefits to patients with cardiovascular disease. The significance of this study is its ability to provide a protective strategy in heart disease in which the mechanism is understood, which is effective, and importantly, which will have the potential to provide savings to the health care system through a reduction in hospital stay.Read moreRead less

Additionally, we have found that drugs and herbicides that block plant chloroplast fat production stop growth of malaria cultures. Type 2 diabetes is a chronic disease which occurs when the pancreas is unable to produce enough insulin for the body to cope with rising blood glucose levels after a meal, and is strongly linked to obesity. We have discovered that fat oversupply activates an enzyme in the pancreas causing defects in insulin release due to glucose. Inhibiting this enzyme helps overcome diabetes, through poorly defined mechanisms that we aim to clarify here. Our work could lead to new therapies for diabetes.
