Cell death is essential for protecting the body against cancer, and defects in cell death pathways contribute to cancer progression. To design new and better cancer therapies we must understand the critical processes which control cell death, and develop effective ways to either reset, or bypass, defects in cell death pathways that contribute to cancer. The program as outlined will elucidate the process of mitochondrial outer membrane permeabilization, a critical event in cell death by apoptosis, and determine how to kill cells in which this event is blocked.Read moreRead less

In particular, Schlafen genes are likely to play a role in inflammatory responses and in blood cell growth. These process clearly have relevance to a range of major human (and animal) diseases including infectious disease, auto-immune disease and leukaemia, and thus a long-term outcome may be improved treatments for such disease. Blood cells are fundamental to health. They play a vital role in maintaining the condition of tissues and organs, fight infections and are essential players in the body's response to injury. Understanding how blood cells are produced and how they function is critical to improving the treatment of disease. With the sequencing of the genome, we now have the tools we need to find the genes controlling these processes. This project will harness the power of modern genetic technologies to dissect the role of novel genes involved in blood cell formation and function, and will open up new therapeutic opportunities for treating the many diseases associated with dysregulation of this important cell system. Structure and function of heterodimeric amino acid transporters. Recently a new family of amino acid transporters has been identified, the members of which are involved in the delivery of amino acids between cells and tissues. All members display a rather unusual structure in that they require two subunits to be functional. The main aim of this study is to investigate the interaction between large and small subunits. The study will give insight how membrane proteins form complexes in the membran ....Structure and function of heterodimeric amino acid transporters. Individual cells communicate with their immediate environment by the process of macropinocytosis, a process that involves the exchange of materials between the extracellular space and a specialised region of the cell termed endosomes. It is an important process in mammalian cells being essential to the correct functioning of many tissues. This project will advance understanding of macropinocytosis at a molecular level. The project is relevant to understanding the functioning of normal cells and the means by which some pathogens can enter cells and also understanding processes involved in tumour progression and metastasis.Read moreRead less

Undertaking large scale projects of this nature requires specialised robotics and dedicated infrastructure for microscopy and tissue preparation. This facility will be the first of its type in Australia will permit researchers to perform genomic scale in situ screens, many as part of large international initiatives in developmental and cellular biology. This large-scale, high-resolution expression profiling infrastructure is required to maintain international competitiveness and will dramatically improve our gene discovery, functional assessment and understanding of vertebrate development.Read moreRead less
Relationship of the functional architecture of the mammalian brain to its microcircuitry. The project seeks to understand a very fundamental issue in neuroscience: how the connectivity and architecture of the cortex are related to the functions of neurones in that area? This will be investigated by imaging the surface of the visual cortex of anaesthetised cats and monkeys as special visual patterns are shown to the eye/s. The overall picture gained of the active and inactive cortical areas will ....Relationship of the functional architecture of the mammalian brain to its microcircuitry. The overall picture gained of the active and inactive cortical areas will be related to the properties of neurones in those areas and to those of individual input and output fibres. An optical imaging equipment will be acquired in 2004 using a recently awarded LIEF grant to the CI.Read moreRead less

Organisation of the cerebral cortex- what, if anything, is a cortical area? Currently, much effort is being directed towards elucidating the roles of different regions of the cerebral cortex. "Maps" of cortical areas have become essential points of reference, exerting a powerful influence on the way we think about the brain. However, is it really appropriate to model the cortex as a mosaic of well-defined areas? This project will challenge the assumptions underlying current brain mapping efforts ....Organisation of the cerebral cortex- what, if anything, is a cortical area? Currently, much effort is being directed towards elucidating the roles of different regions of the cerebral cortex. "Maps" of cortical areas have become essential points of reference, exerting a powerful influence on the way we think about the brain. However, is it really appropriate to model the cortex as a mosaic of well-defined areas? This project will challenge the assumptions underlying current brain mapping efforts. Its main benefits will be the refinement of strategies for brain mapping, including potential for better
application/interpretation of available medical technology, and unique insights on the mechanisms of development and evolution of the nervous system.
