As bacterial superbugs – resistant to multiple antibiotics – dominate the headlines, the pipeline for new antibiotics has all but dried up. High-throughput DNA sequencing heralds a golden opportunity for infectious disease research. By studying the entire collection of genes - the genome - of large numbers of multidrug resistant bacterial strains, we aim to better understand the genetic changes that govern the emergence and global spread of superbugs and translate these findings into the clinic.

Summary
Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS) is an illness affecting up to 240,000 Australians. The cause of ME/CFS is not known, and there are currently no diagnostic tests or effective treatments. Research suggests that ion channels that transfer calcium within cells are dysfunctional in ME/CFS. Our research will investigate ion channels and calcium transfer using immune cells to help develop biomarkers for the illness and discover better treatments for these patients.

Summary
Antibiotic resistance (AR), as highlighted by the WHO, is the most pressing medical need of the 21C – some infections are now untreatable. Our research will focus on the new "superbug" NDM-1 positive E. coli. We will correlate AR and pathogenicity and explore the evolution of these "superbugs" using state-of-the-art sequencing. This research will benefit Australian medicine by predicting timelines of AR epidemics and by conducting the first analyses on the virulence potential of these strains.

Summary
Lung infections are a major disease burden worldwide. Indigenous Australians are affected more; adults with severe lung disease can die in their 30s and 40s. These diseases start early, yet no in-depth studies of bacteria causing lung infections in children have been done. Our expert team proposes to use existing lung fluids from nearly 500 children with chronic lung disease (58% Indigenous), and state-of-the-art laboratory methods, to inform the design of vaccines to prevent lung disease.
