An understanding of how Single minded 1 (SIM1) regulates target genes may allow new pharmaceutical approaches to be designed to combat obesity. As Sim1 belongs to a family of closely related gene regulatory proteins which function in early development and homeostasis, deciphering the molecular control mechanisms of Sim1 may help understand how the related factors function in processes such as angiogenesis, response to low oxygen stress, invasion of environmental pollutants and autism spectrum diseases. The ability to manipulate these factors would be of great benefit in treating a range of disorders, but a thorough molecular understanding of these factors needs be obtained prior to attempting design of pharmaceuticals.Read moreRead less
The Ontogeny of Pain Behaviour: A Novel Neuroimmune Pathway. In Australia, 20% of the population report suffering from chronic pain. In Australia, 20% of the population report suffering from chronic pain. The cost of pain to Australian business per year is over $3 billion. Attempts to explain many chronic pain states, based on current knowledge, have failed. The central hypothesis of this project is that pain sensitivity is determined by programming of the fetal immune system during pregnancy. This research proposal will determine whether exposure to infection early in life determines sensitivity to pain in adult life. In doing so, this research offers a new theoretical explanation for the ontogeny of pain and may begin to account for the many forms of chronic pain that are currently not only unexplainable from current pain theory, but difficult to manage clinically.Read moreRead less

This project will develop technologies for a stem cell therapy platform based on cord blood stem cells, to enable treatment of autoimmune diseases or transplants. Building on the University of Adelaide's frontier demonstration of differentiation of regulatory Thymus (T) cells from cord blood stem cells, the project will develop techniques to expand the numbers of T cells generated. This has the potential to maintain Australia's lead in differentiation of cord blood stem cells and to provide a significant breakthrough in potential treatments of autoimmune diseases (e.g. type 1 diabetes) or transplantation. These diseases affect both a healthy start to life and healthy ageing, and an Australian invention to treat or cure them would have global impact.Read moreRead less
Huntingtin-associated protein 1 controls cell communication. The purpose of this study is to identify the mechanisms by which a novel regulator of cell communication which we have identified is able to control the release of chemical signals from a cell. This project will provide critical insight into a cellular pathway that underlies hormone secretion, neurotransmission and higher brain functions.
Function and redundancy of SOX genes in the mammalian sex determination pathway. We are studying a mouse model of abnormal sex organ development in which genetically female mice develop as males. Our basic research program will lead to greater understanding of the genetic switch controlling the formation of male and female characteristics. This research should in turn provide insight into the causes of defects in patients with disorders of sex development, helping to inform the difficult clinica ....Function and redundancy of SOX genes in the mammalian sex determination pathway. We are studying a mouse model of abnormal sex organ development in which genetically female mice develop as males. Our basic research program will lead to greater understanding of the genetic switch controlling the formation of male and female characteristics. This research should in turn provide insight into the causes of defects in patients with disorders of sex development, helping to inform the difficult clinical decisions that need to be made for their treatment, and ultimately leading to better management and therapeutic strategies. Our studies may also provide unique methods to control the exotic mouse population, using the daughterless strategy.Read moreRead less

Yet, currently the understanding of how circRNAs regulate biological processes is very poor. This project aims to use pooled shRNA libraries to screen a large panel of cell lines and systematically identify cellular activities that are regulated by circRNAs. The expected outcome of this study will be a catalogue of functionally active circRNAs. Over the past decades, the wealth of knowledge on the function of linear mRNAs has had a significant impact on medicine and agriculture. Similarly understanding how circRNAs regulate cellular activities may have an analogous impact on humans.Read moreRead less

Medical Device technologies embrace a wide range of scientific, engineering and medical knowledge, with the goal of assisting a clinical professional (doctor or nurse) deliver a service to a patient in an efficacious, cost effective manner. Development of appropriate medical devices, whether for diagnosis, treatment or prevention of disease or disability, is critical to improving health care and reducing health care costs. To be successful, a device must include all relevant disciplines in the research, development and testing phases. This network will bring together these groups, promoting knowledge sharing and cross-disciplinary investigations that illuminate current device limitations and potential solutions.Read moreRead less

The femur is a light-weight structure designed to best perform in life. However, the complex tissue architecture, microstructural organisation and its complex loading regimens make it difficult to understand how the femur can deform and fracture. This project studies femoral fractures by modelling the proximal femur with a micrometric level of detail. Synchrotron femur images are taken in loaded and unloaded conditions. Cortical strain and fracture are measured, replicating possible multiaxial loads. Micro finite-element models will be used to study the contribution that the bone tissue architecture, tissue structure and activity types make to the fracture. The resulting knowledge will have future orthopaedic applications.Read moreRead less
