Development of Liposaccharide Peptide Conjugates for Peptide Drug Delivery. We aim to adapt the Solid Phase Oligosaccharide Synthesis technology patented by Alchemia to develop methodologies that will allow the assembly of sugar, lipid and peptide units on a single solid phase resin. This synthetic technology will then be used to generate a library of liposaccharide conjugates of two model peptides (LHRH and TRH) to investigate and optimise a broadly applicable peptide drug delivery system whic ....Development of Liposaccharide Peptide Conjugates for Peptide Drug Delivery. A combinatorial approach to creation of new epitopes with biological activity. We aim to develop and extend the Mosaic technology patented by Proxima Concepts Ltd. as a new methodolgy for dynamic combinatorial chemistry and drug delivery. The technology relies on bringing chemical structures of interest in close proximity by presenting them on the surface of a micelle type structure. These structures will also be developed at a targeted drug delivery vehicle. The technology will impact on the ....A combinatorial approach to creation of new epitopes with biological activity. BIOCATALYSTS MINED FROM CYTOCHROME P450 LIBRARIES: AN INNOVATIVE TOOL FOR ACCELERATING DRUG DEVELOPMENT. The cytochrome P450s (P450s) are a family of enzymes that are perhaps the most versatile biological catalysts known. DNA shuffling is an emerging technique that takes the genes encoding families of enzymes and creates libraries of catalysts with both improved and novel properties. We will obtain proof of concept that shuffled P450 libraries can be screened and optimized for use as biocatalys ....BIOCATALYSTS MINED FROM CYTOCHROME P450 LIBRARIES: AN INNOVATIVE TOOL FOR ACCELERATING DRUG DEVELOPMENT. DNA shuffling is an emerging technique that takes the genes encoding families of enzymes and creates libraries of catalysts with both improved and novel properties. We will obtain proof of concept that shuffled P450 libraries can be screened and optimized for use as biocatalysts in drug development. The methodologies developed here will overcome two critical bottlenecks in current drug development: the optimisation and metabolic profiling of new drug candidates. This will yield important benefits in accelerating the optimisation and safety testing of drugs under development.Read moreRead less

It is estimated that chronic pain affects one in five individuals in the general community in Australia and worldwide, with prevalence rates correlated directly with advancing age. Chronic pain not only adversely affects the quality of life of individuals but it also places a large economic burden on our healthcare system. Development and validation of an in vitro method to successfully identify novel morphine-like strong analgesics with a reduced propensity for producing respiratory depression or constipation, has the potential to not only improve pain management for individuals and to reduce the economic burden of chronic pain on the Australian healthcare system, but it is also likely to produce direct economic benefits to our nation. Topical peptide delivery for cosmetic and therapeutic benefits. Milk is a major Australian agricultural commodity and is now used in a number of topical products for the management of various skin conditions including chafing in babies, eczema and ageing skin. Hence, this work hopes to contribute to promoting and maintaining good health of Australians.
In addition, there is considerable research being conducted on peptide development for a range of diseases and there may be a possibility of ....Topical peptide delivery for cosmetic and therapeutic benefits. Milk is a major Australian agricultural commodity and is now used in a number of topical products for the management of various skin conditions including chafing in babies, eczema and ageing skin. Hence, this work hopes to contribute to promoting and maintaining good health of Australians.
In addition, there is considerable research being conducted on peptide development for a range of diseases and there may be a possibility of delivering these by the skin.
This work, in seeking to understand some of the fundamental determinants governing how exogenously applied peptides distribute in the skin, is also contributing to the development of Australian pharmaceutical and cosmetic industries.Read moreRead less
Targeting virulence of Pseudomonas aeruginosa by inhibiting oxidative protein folding. Our research will lead to the development of compounds with a novel anti-virulence/antibacterial mode of action for further drug development.
Novel polymeric microparticles for slow-release intrathecal delivery of analgesics. Unrelieved chronic pain is a large area of unmet medical need, adversely affecting the lives of millions of people worldwide. This project seeks to develop innovative methods for delivering a novel pain-relieving medication into the spinal fluid in close proximity to the cells that transmit pain messages to the brain. The goal is to develop a new treatment to produce prolonged periods of pain relief in those pat ....Novel polymeric microparticles for slow-release intrathecal delivery of analgesics. The goal is to develop a new treatment to produce prolonged periods of pain relief in those patients who do not obtain benefit from existing pain medications. The technology that will be developed has the potential to be applied to a broad range of analgesic drug treatments.Read moreRead less
Relationship between melanosome distribution and skin colour. This work seeks to examine how the colour of our skin is related to the distribution of melanosomes. This work may allow us to better understand how to develop novel therapies that may alter skin colour and provide protection from the sun (frontier technologies) but may also, in the longer term, lead to healthier approaches to skin management - especially for ageing skin.
Diversity of Salinispora actinobacteria producing pharmaceutically relevant natural products from Australian marine sponges. By investigating the distribution of marine microbial resources relevant to drug discovery, we will directly contribute to ARC's Research Priority I - An Environmentally Sustainable Australia Priority Goal and the Priority Goal 'Sustainable use of Australia's biodiversity'. We will determine sources of marine bacteria and their genes useful for discovery of new natural products for treatment of human diseases. We will do this by understanding where new strains of Salinispora bacteria may be isolated and how they are distributed in association with Australian marine sponge fauna, and by determining the distribution and chemical and genetic diversity of novel marine Salinispora bacteria.Read moreRead less
