Summary
Our proposal will provide a deep mechanistic framework to inform both clinicians in diagnosis and management of RUVBL related diseases and also therapeutically, as industry looks to use these proteins as drug targets. The great excitement of RUVBL in translation has outpaced the gathering of vital knowledge underpinning the function; knowledge this proposal will provide for the first time.
The molecular interactome and functions of circular RNAs. This project aims to identify the functions of circular RNAs, the most contemporary and enigmatic family of RNA molecules. While their abundance suggests they are important, it is unclear how they function at the molecular level. Making muscle: molecular dissection of membrane domain formation. For a muscle to contract efficiently in response to an electrical signal it requires the formation of an extensive system of hollow membranous tubules through which the signal can be propagated. Protein phosphorylation is a dynamic process regulated by both protein kinases and protein phosphatases. Characterisation of p14ARF intracellular trafficking pathways. Over 3500 new cases of melanoma are diagnosed in NSW each year, and one of the most important proteins involved in suppressing melanoma initiation or growth is p14ARF. This project will characterise the movement and functions of this protein with the aim of identifying novel targets for more effective drug therapies.
Understanding how Plasmepsin V directs export of malaria virulence proteins to the host cell. This project aims to characterise how malaria parasites survive and manipulate infected host cells by exporting virulence proteins. This project may identify essential proteins that allow the malaria parasite to transform the host in order to survive, replicate and hide from the immune system and provide new data on protein export in liver-stages.
Control of developmental switches by importin 5. Aims: This project will study a key molecular switch called IPO5, a protein that is required for cells and organs to form and function normally, and it will reveal how it works.
Significance: These experiments will provide the first complete description of how this molecular switch controls the behaviour of a cell across its lifespan. IPO5 is highly conserved, so these studies will be relevant to a wide range of animals.

This will help address a major issue in the field: How do interactions between transcription factors regulate DNA-binding specificity. To ensure competitive growth and survival, plant and animal cells have sophisticated mechanisms of information transfer. One such process is efficient export of molecules from the cell nucleus (the coding space) to the cell cytoplasm (the protein synthesis space).
