Summary
Understanding the genes that underpin the immune system is vital for studying the causes and effects of diseases. In particular, autoimmune diseases are strongly associated with an individual having certain genetic types. We plan to develop methods to type immune-related genes very cheaply so they can be studied in very large samples. We will apply the methods we develop to genetic studies of psoriasis and multiple sclerosis, and make data and methods available to other scientists.

Psoriatic arthritis is a chronic joint disease which is linked with the skin condition called psoriasis. People who have psoriatic arthritis can develop deformities in their joints and suffer from decreased quality of life. Psoriatic arthritis can present in a wide range of ways, but the reasons for this are not understood. This research aims to explain some of this variation and to predict which patients will develop more severe disease so treatment can be targeted to improve their outcome.

To achieve an accurate molecular understanding of innate immune system receptor signalling, both intracellularly and in whole organisms, in health and disease. This knowledge will then be used to generate better treatments for the extensive range of human diseases that are caused or exacerbated by dysfunctional innate immune signalling, including Crohn's disease, psoriasis and cancer.

Many products are applied to the skin to prevent skin cancer or to treat skin diseases. This project seeks to better understand how we can make such products more affective, safer and appropriate for conditions such as psoriasis. One major component of the grant is concerned with the evaluation of nanotechnology products applied to the skin.

Summary
Autoimmune diseases, including rheumatoid arthritis and psoriasis, have profound impacts on the lives of many Australians. New drugs are required for these diseases as existing treatments are expensive, become less effective with repeated use or cause adverse side effects. My project will work towards addressing this need by investigating the potential of ultrastable cyclic miniproteins as scaffolds for displaying bioactive peptides that are known to restore immune self-tolerance.
